摘  要:吗啡常用于慢性疼痛的治疗,但其临床应用受限于其药学特性,如不良反应较多、成瘾风险较高。吗啡成瘾涉及脑源性神经营养因子(BDNF)及其信号通路在内的多条通路。BDNF及其下游信号通路广泛参与脑神经元的功能,涉及多种精神疾病。慢性吗啡暴露可引起脑及血浆中BDNF表达的变化,同时可引起脑中BDNF及其下游信号磷脂酶Cγ、磷脂酰肌醇-3-激酶和胞外信号调节激酶表达的变化,且外源性BDNF对吗啡成瘾具有保护作用。未来需进一步发掘BDNF及其通路与吗啡成瘾的关系,以为改善吗啡成瘾提供指导性建议。Morphine is commonly used for the treatment of chronic pain,but its clinical use is limited by its pharmaceutical characteristics,such as more side effects and a higher risk of addiction.Morphine addiction involves many pathways including brain-derived neurotrophic factor(BDNF)and its signaling pathway.BDNF and its downstream signaling pathways are widely involved in the function of brain neurons,and are involved in a variety of mental diseases.Chronic morphine exposure can induce changes in the expression of BDNF in brain and plasma,as well as changes in the expression of BDNF and its downstream signals phospholipase Cγ,phosphatidylinositol-3-kinase and extracellular signal-regulated kinase in brain.Moreover,exogenous BDNF has a protective effect on morphine addiction.In the future,the relationship between BDNF and its pathway and morphine addiction should be further explored to provide guidance for improving morphine addiction.
