摘 要:经典Wnt信号通路是调控胚胎发育、组织稳态和损伤修复的重要通路,在进化过程中高度保守,但其信号转导异常也与恶性肿瘤的发生、进展、耐药及复发等密切相关。经典Wnt信号通路过度激活通过影响免疫细胞诱导肿瘤免疫耐受并抑制T细胞浸润到肿瘤微环境,破坏正常的免疫监测,导致恶性肿瘤患者对免疫检查点抑制剂产生耐药。经典Wnt信号通路调节剂能够增强肿瘤免疫治疗的效果。因此,进一步阐明经典Wnt信号通路在肿瘤免疫耐受发生中的作用及机制将为恶性肿瘤的治疗提供更多策略。The canonical Wnt signaling pathway is an important pathway regulating embryonic development,tissue homeostasis and injury repair.It is highly conserved in the evolutionary process,while its abnormal signal transduction is also closely related to the occurrence,progression,drug resistance and recurrence of cancers.The overactivation of canonical Wnt signaling pathway induces tumor immune tolerance by influencing immune cells and inhibiting T cell infiltration into tumor microenvironment,thus disrupting normal immune surveillance and leading to drug resistance to immune checkpoint inhibitors in the patients with cancer.Canonical Wnt signaling pathway modulators have been found to enhance the effect of tumor immunotherapy.Therefore,further elucidating the role and mechanism of canonical Wnt signaling pathway in the development of tumor immune tolerance will provide more strategies for the treatment of cancer.
