摘 要:本试验旨在探究脂蛋白分解酶(lipoprotein lipase,LPL)基因在水牛不同组织及泌乳期乳腺组织的表达情况,为今后研究该基因在奶水牛泌乳方面的作用提供参考。运用生物信息学方法以GenBank中黄牛LPL基因序列为模板成功克隆了水牛LPL基因完整编码序列(CDS),并对其序列进行分析,同时使用实时荧光定量PCR技术检测LPL在水牛不同组织和不同泌乳时期中的转录水平。结果显示,水牛LPL基因CDS序列长度为1437bp,水牛LPL亚细胞定位于细胞外(包括细胞壁)、线粒体、内质网和空泡的比例分别为55.6%、22.2%、11.1%和11.1%,说明水牛LPL主要分布在细胞外;信号肽分析结果显示,该蛋白信号肽的切割位点位于SRGGL之间,概率为0.4029;组织表达分析结果表明,水牛LPL基因在乳腺的表达量最高(P<0.01),其他组织的表达量从高到低依次为心、输卵管、肺、脾、肾、卵巢、大肠、淋巴、子宫、胃、肝、大脑和垂体;泌乳期表达分析结果表明,LPL在D50时的乳腺组织中表达量最高(P<0.01),推测LPL在水牛泌乳盛期起重要作用。In order to understand the mRNA expression of lipoprotein lipase in different tissues and lactation period of buffalo and,providing a basis for future studies on the role of this gene in milk buffalo lactation.The buffalo LPL(lipoprotein lipase)gene was cloned and analyzed by bioinformatics method in this study,and the qPCR was used to detect the transcription level of LPL in different tissues and mammary gland during lactation of buffalo.The results showed that the CDS sequence length of buffalo LPL gene was 1437 bp,and it could encode 478 amino acids.Subcellular localization of LPL was in the extracell,including cell wall(55.6%),mitochondrial(22.2%),endoplasmic reticulum(11.1%),and vacuolar(11.1%),mainly in the extracell.There was a signal peptide located between SRG-GL but no transmembrane domain.The qPCR results showed that the expression of buffalo LPL gene in the mammary gland was significantly higher than other tissues(P<0.01).The results of lactation expression showed that the expression of LPL was highest in mammary gland at D50(P<0.01),it was suggesting LPL played an important role in the peak lactation of buffalo.
