摘 要:【目的】快速扩繁长寿花优良品种,建立长寿花叶片离体组织繁殖体系。【方法】以长寿花幼嫩叶片为外植体,考察灭菌方法对叶片再生能力的影响,比较培养基中激素NAA和6-BA的质量浓度对长寿花叶片愈伤组织的诱导及不定芽生根的影响。【结果】长寿花幼嫩叶片的最佳消毒方法为2%次氯酸钠溶液消毒10 min,无菌水冲洗4次,75%乙醇消毒30 s,再次用无菌水冲洗4次后灭菌完成。最适愈伤组织诱导培养基是MS+6-BA 0.2 mg/L+NAA 2 mg/L,愈伤组织诱导率达95.6%;最适分化培养基是MS+NAA 0.2 mg/L+6-BA 1.0 mg/L,丛芽分化率达90.0%;最适生根培养基是1/2 MS+NAA 0.5 mg/L,生根率达96.7%,经炼苗成活率高达90%以上。【结论】该研究建立长寿花组织培养的再生体系,诱导率、出芽率、生根率及移栽成活率均达到90%以上,显著缩短移栽时间,有效降低繁殖成本。【Objective】To product excellent Kalanchoe blossfeldiana rapidly,the tissue culture system was established.【Method】Kalanchoe blossfeldiana leaves were used for explants to study the regeneration ability of the leaves with different sterilization methods,the effect of hormones(NAA or 6-BA)with various concentrations in the culture medium on callus inducing,shoot differentiation,and rooting.【Result】The best sterilization method was sterilizing the tissue in 2%NaClO for 10 min,washing with disinfection water for 4 times,then in 75%ethanol for 30 s,washing 4 times with disinfection water.The results showed that MS with 6-BA 0.2 mg/L+NAA 2 mg/L was suited for the best callus induction medium.The callus induction rate was 95.6%.MS with NAA 0.2 mg/L+6-BA 1.0 mg/L was the best suited for adventitious bud differentiation and the bud differentiation rate was 90.0%.1/2 MS with NAA 0.5 mg/L was the best suited for root formation.The rooting rate was 96.7%and the seeding survival rate was more than 90%.【Conclusion】The regeneration system of tissue culture of Kalanchoe blossfeldiana was established in this study.The induction rate,budding rate,rooting rate and transplanting survival rate were more than 90%,which significantly shortened the transplanting time and effectively reduced the propagation cost.
