鲢鱼感染嗜水气单胞菌FAH5后免疫因子的变化研究

摘  要:为研究鲢鱼受到嗜水气单胞菌FAH5感染后机体免疫因子的变化过程,在健康鲢鱼体内注射嗜水气单胞菌FAH5,感染后12~108 h收集鲢鱼的血液和肝脏,使用分光光度计测定血液铁浓度和载体蛋白浓度,使用ICP-AES测定肝脏铁浓度,使用实时荧光定量PCR测定铁调素基因hepc20、白细胞介素基因il-1、酪氨酸激酶基因jak1、应激因子基因stat1的表达量。结果表明,健康鲢鱼受到嗜水气单胞菌FAH5感染不同时间后,鲢鱼血液铁浓度出现不同程度的降低,其中24和36 h时与对照组比较,达到显著差异;血液载铁蛋白浓度与对照组比较,出现不同程度上升,但均未达到显著差异;肝脏铁浓度与对照组比较,出现不同程度上升,但均未达到显著差异。肝脏中hepc20铁调素基因的表达量在12和24 h时与对照组比较上升,达到显著差异;肝脏中il-1白细胞介素基因的表达量在24和36 h时与对照组比较上升,达到显著差异;肝脏中酪氨酸激酶jak1基因的表达量在12和24 h时与对照组比较上升,达到显著差异;肝脏中stat1应激因子基因的表达量在12和24 h时与对照组比较上升,达到显著差异。因此,鲢鱼机体通过调节铁调素基因hepc20、白细胞介素基因il-1、酪氨酸激酶基因jak1、应激因子基因stat1等免疫基因的表达的变化,降低机体游离铁浓度和增加储存铁浓度来应对嗜水气单胞菌FAH5的感染。In order to study the change process of immune factors of Silver carp infected with Aeromonas hydrophila FAH5,after injecting Aeromonas hydrophila FAH5 into healthy Silver carp,the blood and liver of Silver carp were collected 12-108 h after infection,the blood iron concentration and blood ferritin concentration were measured by spectrophotometer,and the liver iron concentration was measured by ICP-AES.The expressions of hepc20,il-1,jak1 and stat1 were measured by real-time fluorescence quantitative PCR.After healthy Silver carp were infected with Aeromonas hydrophila FAH5 for different times,the blood iron concentration of Silver carp decreased in varying degrees,and there was significant difference compared with the control group at 24 and 36 hours.Compared with the control group,the blood ferritin concentration increased in varying degrees,but there was no significant difference.Compared with the control group,liver iron concentration increased in varying degrees,but there was no significant difference.The expression of hepc20 ferritin gene in liver increased significantly compared with the control group at 12 and 24 hours.The expression of il-1 interleukin gene in liver increased significantly compared with the control group at 24 and 36 hours;the expression of tyrosine kinase jak1 gene in liver increased significantly compared with the control group at 12 and 24 hours;the expression of stat1 stress factor gene in liver increased significantly compared with the control group at 12 and 24 hours.Therefore,the Silver carp body responds to the infection of Aeromonas hydrophila FAH5 by regulating the expression of immune genes such as hepc20,il-1,jak1 and stat1,reducing the free iron concentration and increasing the stored iron concentration.
