摘 要:选取从江香猪附睾组织作为试验对象,运用免疫组织化学检测结合Western blot分析3β-羟基甾脱氢酶(3β-HSD)在初情期前、初情期、初情期后、性成熟期4个发育时期从江香猪附睾中的表达情况,探究3β-HSD在哺乳动物附睾中发挥的生理作用.免疫组织化学检测显示,从江香猪附睾3β-HSD在4个发育时期均有分布,其表达主要集中在附睾管上皮周围的平滑肌和管腔微绒毛上.Western blot分析显示,初情期3β-HSD的表达量极显著高于初情期前、初情期后、性成熟期(P<0.01),而初情期前、初情期后、性成熟期的表达量两两之间并无统计学差异(P>0.05).以上结果表明,3β-HSD主要定位于从江香猪附睾平滑肌和微绒毛上,且呈年龄依赖性表达,初情期的表达量最高,推测3β-HSD参与了初情期附睾功能的调节.In order to elucidate the physiological role of 3β-hydroxysteroid dehydrogenase(3β-HSD) in the mammalian epididymis, the expression pattern of 3β-HSD was investigated during 4 developmental phases of epididymis(pre-puberty, puberty, post-puberty and sexual maturity) in Congjiang Xiang pig by immunohistochemistry combined with Western blotting. The immunohistochemical results showed that 3β-HSD existed in the epididymal tissues in all the 4 developmental phases, and was mainly distributed in the smooth muscle around epididymal epithelium and the microvilli of lumen. Western blotting showed that the expression of 3β-HSD in puberty was significantly higher than the expressions in the other 3 phases(P<0.01), while its expressions in the 3 phases were not significantly different(P>0.05). In conclusion, 3β-HSD was mainly concentrated in the smooth muscle around epididymal epithelium and the microvilli of lumen of Congjiang Xiang pig, and expressed in an age-dependent manner with the highest expression during the puberty, indicating the involvement of 3β-HSD in the regulation of epididymal function during peri-puberty.
