摘  要:目的:探讨嗜粘蛋白阿克曼菌(AKK菌)对高脂饲料诱导的肥胖SD大鼠糖脂代谢相关指标的影响。方法:健康SPF级雄性SD大鼠,选取10只作为对照组,喂普通饲料;高脂饲料诱导肥胖大鼠模型,选取成功复制的肥胖大鼠20只,随机分为肥胖模型组和AKK菌干预组。对照组灌胃生理盐水,肥胖模型组灌胃不含AKK菌的混悬液,AKK菌干预组灌胃含1×10^(9)CFU AKK菌的混悬液。每日灌胃1次,每次1 mL/kg。灌胃4 w后,3组动物分别测定血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)浓度,进行糖耐量试验[(空腹血糖(FPG)、餐后0.5 h血糖(0.5 hPG)、餐后1 h血糖(1 hPG)、餐后2 h血糖(2 hPG)],测定血清胰岛素水平及空腹体重、肝脏重量及附睾脂肪重量;HE染色观察大鼠肝脏脂肪浸润情况及附睾脂肪细胞脂滴大小。结果:AKK菌灌胃4 w后,与肥胖模型组相比,大鼠肝脏重、肝指数(%)、FPG、1 hPG、2 hPG、血糖曲线下面积、TG、TC、LDL-C、AST、ALT水平均明显下降,差异有统计学意义(P<0.05);附睾脂肪重、体脂百分比、0.5 hPG和胰岛素水平均有下降趋势,但差异无统计学意义(P>0.05);HDL-C有升高趋势,但差异无统计学意义(P>0.05);HE染色观察显示,肝脏组织的脂肪浸润程度以及附睾脂肪细胞的脂滴面积均小于肥胖组模型组。结论:AKK菌可显著降低高脂饲料诱导的肥胖大鼠的体重、血糖血脂、转氨酶水平,可能通过提高大鼠胰岛素敏感性,改善其肝功能。Objective To investigate the effects of Akkermansia muciniphila(AKK)on glucose and lipid metabolism in obese SD rats fed with high-fat diet.Method Totally 10 healthy SPF male SD rats were selected as the control group and fed with ordinary diet.The obese rat model induced by high-fat diet,20 successfully replicated obese rats were randomly divided into obesity model group and AKK bacteria intervention group,and were respectively fed with normal saline,the suspension without AKK bacteria,the suspension containing AKK bacteria the dose was 1×10^(9)CFU respectively once a day,1 mL/kg each time.After gavage for 4 weeks,the levels of serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),triglyceride(TG),total cholesterol(TC),high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C),glucose tolerance test[fasting blood glucose(FPG),half hour postprandial blood glucose(0.5 hPG),1 h postprandial blood glucose(1hPG),2 h postprandial blood glucose(2 hPG)],insulin level,fasting weight,liver weight and epididymal fat weight were measured in the three groups,and HE staining was performed in order to observe the fat accumulation in liver and the size of epididymal adipocytes.Result After given the AKK bacteria for four weeks,compared with the obesity model group,the liver weight,liver index(%),FPG,1 hPG,2 hPG,GAUS,TG,TC,LDL-C,AST and ALT levels decreased significantly(P<0.05).The epididymal fat weight,body fat percentage,0.5 hPG and insulin levels all decreased,but the difference was not statistically significant(P>0.05).HDL-C increased,but the difference was not statistically significant(P>0.05).HE staining showed that AKK bacteria significantly reduce the fat accumulation in liver and the size of epididymal adipocytes compared with the obese rats.Conclusion AKK bacteria can significantly reduce the body weight,blood glucose,blood lipid and transaminase levels of obese rats induced by high-fat diet,which may improve their liver function by increasing insulin sensitivity.
