摘　　要：目的探讨PTPN12低表达参与调控肺癌H1299细胞凋亡的作用及对放疗敏感性的影响。方法选取肺癌患者82例,应用QIAamp DNA FFPE Tissue Kit试剂盒进行样本DNA提取,采用NanoDrop1000 spectrophotometer测定样本DNA浓度与纯度,所提取的DNA采用紫外分光光度仪进行浓度检测与质量评估。采用化学发光法检测外周血血清肿瘤标志物[癌胚抗原(CEA)、糖类抗原125(CA125)、细胞角蛋白19片段(CYFRA21-1)、鳞状上皮细胞癌抗原(SCC-Ag)、神经元特异性烯醇化酶(NSE)、肿瘤特异性生长因子(TSGF)]。扩增产物采用western blot法进行蛋白产物的定性和定量分析。结果PTPN12表达对肺癌病情的调控作用分析结果可见,在组织分化和病理分期更严重的患者中PTPN12-mRNA表达更低,且在随访生存期更低的患者中也可见表达更低,均有统计学意义;但PTPN12蛋白表达只在不同病理分期患者中检出差别有统计学意义,且同mRNA表达规律相一致;而在不同组织分化和预后患者间差别尚未见统计学意义,与样本量等因素限制有关。PTPN12基因在肺癌组织中突变38例,突变患者血清CEA高于野生型患者,CA125、CYFRA21-1低于野生型患者(P均<0.05)。同时,PTPN12低表达与NSCLC患者的放射敏感性密切相关,而且PTPN12基因下调能明显增加H1299细胞的放射敏感性。结论PTPN12低表达同肺癌患者的放射敏感性存在关联,PTPN12下调可以有效增加H1299细胞的放射敏感性,可以考虑作为提示患者的放射敏感性的靶点,并为靶向治疗联合放疗提供更多依据。Objective To investigate the role of low expression of ptpn12 in the regulation of apoptosis of lung cancer H1299 cells and its effect on radiosensitivity.Methods 82 patients with lung cancer were selected,and the sample DNA was extracted with qiaamp DNA FFPE tissue kit.The concentration and purity of the sample DNA were determined by nanodrop1000 spectrophotometer.The extracted DNA was detected by UV spectrophotometer and its quality was evaluated.Serum tumor markers[carcinoembryonic antigen(CEA),carbohydrate antigen 125(CA125),cytokeratin 19 fragment(CYFRA21-1),squamous cell carcinoma antigen(SCC Ag),neuron specific enolase(NSE),tumor specific growth factor(TSGF)]were detected by chemiluminescence.The amplified products were analyzed qualitatively and quantitatively by Western blot.Results The analysis of the regulatory effect of ptpn12 expression on the condition of lung cancer showed that the expression of ptpn12 mRNA was lower in patients with more severe tissue differentiation and disease stage,and it was also lower in patients with lower follow-up survival,which was statistically significant;However,the difference in ptpn12 protein expression was only detected in patients with different pathological stages,and the difference was statistically significant,which was consistent with the rule of mRNA expression;However,there was no significant difference in patients with different histological differentiation,which was related to the limitation of sample size and other factors.ptpn12 in lung cancer tissues had 38 mutation cases,serum CEA in mutation patients was higher than wild type cases,while CA125,CYFRA21-1 were lower than wild type cases(P<0.05).At the same time,the low expression of ptpn12 was closely related to the radiosensitivity of NSCLC patients,and the down-regulation of ptpn12 gene can significantly increase the radiosensitivity of H1299 cells.Conclusion The low expression of ptpn12 is closely related to the radiosensitivity of NSCLC patients,and the down-regulation of ptpn12 gene can significantl
