摘  要:目的:探究细胞黏附分子(CD44v6)在骨肉瘤组织中的表达及沉默CD44v6对骨肉瘤细胞增殖和转移的影响。方法:实时荧光定量PCR(qRT-PCR)和蛋白免疫印迹法(Western Blot)检测骨肉瘤组织和细胞中CD44v6表达水平。将骨肉瘤细胞U2OS分为空白对照组(Control,空白培养细胞)、si-NC组(转染si-NC)、si-CD44v6组(转染si-CD44v6);qRT-PCR检测CD44v6 mRNA表达水平;Western Blot检测CD44v6、增殖细胞核抗原(PCNA)、细胞增殖核抗原67(Ki67)、钙黏附蛋白E(E-cadherin)、钙黏附蛋白N(N-cadherin)、基质金属蛋白酶9(MMP9)蛋白表达;细胞计数试剂盒(CCK8)、克隆形成实验检测细胞增殖;Transwell检测细胞迁移、侵袭。结果:骨肉瘤组织和细胞(U2OS、MG63)中CD44v6 mRNA、蛋白表达水平明显高于癌旁组织和成骨细胞hFOB1.19。si-CD44v6组骨肉瘤细胞中CD44v6 mRNA和蛋白表达水平、PCNA、Ki67、N-cadherin、MMP9蛋白表达低于Control组、si-NC组,E-cadherin蛋白表达高于Control组、si-NC组,克隆细胞数目、迁移细胞数目、侵袭细胞数目少于Control组、si-NC组;其中Control组、si-NC组骨肉瘤细胞中各指标差异不显著。结论:在骨肉瘤组织和细胞内CD44v6高表达,沉默CD44v6可抑制骨肉瘤细胞增殖、迁移、侵袭和上皮间质转化。Objective:To explore the expression of cell adhesion molecule(CD44 v6)in osteosarcoma tissues and the efficacy of silencing CD44 v6 on the proliferation and metastasis of osteosarcoma cells.Methods:Real-time fluorescent quantitative PCR(qRT-PCR)and Western Blot were used to detect the expression level of CD44 v6 in osteosarcoma tissues and cells.Taking the osteosarcoma cell U2 OS as the research object,the cells were divided into blank control group(Control,blank cultured cells),si-NC group(transfected with si-NC)and si-CD44 v6 group(transfected with si-CD44 v6).The expression level of CD44 v6 mRNA was detected by qRT-PCR;Western Blot was used to detect the expression of CD44 v6,proliferating cell nuclear antigen(PCNA),cell proliferating nuclear antigen 67(Ki67),cadherin E(E-cadherin),cadherin N(N-cadherin)and matrix metalloproteinase 9(MMP9)Protein;cell counting kit(CCK8)and clone formation test were used to detect cell proliferation.Transwell was used to detect cell migration and invasion.Results:The expression levels of CD44 v6 mRNA,protein in osteosarcoma tissues and cells(U2 OS,MG63)were significantly higher than that in adjacent tissues and osteoblasts hFOB1.19.The expression levels of CD44 v6 mRNA,protein,PCNA,Ki67,N-cadheri and MMP9 protein in osteosarcoma cells in the si-CD44 v6 group were lower than that in the control and si-NC groups,and the E-cadherin protein expression was higher than those in the control and si-NC groups.The number of cloned cells,migrating cells and invasive cells were less than that of the control group and si-NC group.Among them,there was no significant difference in the indexes of osteosarcoma cells in the control group and si-NC group.Conclusion:The CD44 v6 is highly expressed in osteosarcoma tissues and cells.The silence of CD44 v6 can inhibit the proliferation,migration,invasion and epithelial-mesenchymal transition of osteosarcoma cells.
