摘  要:目的研究GRK5-Moesin通路在胶质瘤中的分布特点及作用。方法采用慢病毒转染法制备GRK5上调/下调的胶质瘤细胞系,Western blot和qRT-PCR鉴定转染效果,Western blot分析GRK5改变对Moesin表达的影响。免疫荧光分析GRK5与Moesin在胶质瘤组织中的亚细胞定位及分布特征。采用CCK-8法、细胞划痕法、Transwell法和流式细胞术分别检测U87细胞增殖、迁移、侵袭和凋亡情况。结果GRK5、Moesin及CD44共定位于胶质母细胞瘤的细胞膜。GRK5-Moesin-CD44在胶质瘤壁龛结构中富集表达。GRK5-Moesin与胶质瘤血管关系密切,但各血管中的GRK5和Moesin分布位置存在差异。靶向GRK5-Moesin可影响U87胶质瘤细胞生物学活性。结论GRK5-Moesin可促进胶质母细胞瘤的恶性进展,并且与胶质瘤干细胞关系密切。Objective To investigate the distribution characteristics and role of GRK5-Moesin pathway in glioma. Methods Glioma cell lines with GRK5 up-regulated/down-regulated were prepared by lentivirus transfection, which were identified by Western blot and qRT-PCR. Effect of GRK5 changes on Moesin expression was detected by Western blot. Immunofluorescence analyzed subcellular localization and distribution characteristics of GRK5 and Moesin in glioma tissues. Proliferation, migration, invasion and apoptosis of U87 cells were detected by CCK-8 assay, Cell scratch assay, Transwell assay and flow cytometry, respectively. Results GRK5, Moesin and CD44 were co-located in glioblastoma cell membrane. GRK5-Moesin were enriched in glioma niches and glioma blood vessels, but the distribution of GRK5 and Moesin in each blood vessel was different. Targeting GRK5-Moesin could affect the biological activity of U87 glioma cells. Conclusion GRK5-Moesin can promote the malignant progression of glioma and is closely related to glioma stem cells.
