摘　　要：目的研究LAMP5在结直肠癌组织中的表达水平及其与临床预后相关性,探讨其在结直肠癌发生发展中的作用。方法使用GEPIA和LinkedOmics数据库分析LAMP5在结直肠癌组织和配对癌旁组织中的表达水平、临床参数结直肠癌患者生存预后的关系并通过临床样本验证。使用LinkedOmics数据库和DAVID网站进行基因富集分析,研究LAMP5在结直肠癌发生发展中涉及的分子功能和相关信号通路。结果免疫组化结果显示,LAMP5在结直肠癌组织中的表达水平高于配对癌旁组织(n=120,P<0.05);LAMP5表达水平与结直肠癌分化程度呈负相关,与N分期和M分期呈正相关(P<0.05),但与年龄、性别和T分期无相关性。GEPIA分析和临床样本Kaplan-Meier分析显示LAMP5高表达结直肠癌患者的总生存期和无病生存期均短于LAMP5低表达者(P<0.05),COX回归分析提示LAMP5高表达是结直肠癌患者独立预后因素。GO分析显示LAMP5与结直肠癌细胞外基质、细胞外基质黏附、细胞外基质定位和胶原定位等功能相关(P<0.05);KEGG分析显示LAMP5可能与TGF-β和PI3K-AKT通路相关(P<0.05)。结论LAMP5在结直肠癌组织中表达水平升高,并且与结直肠癌分化水平、淋巴结转移、远处转移和临床预后相关,因此LAMP5可能作为结直肠癌诊断、治疗和预后预测新的分子指标。Objective To investigate the expression of LAMP5 in colorectal cancer(CRC)and the relationship between the expression of LAMP5 and prognosis of patients with colorectal cancer,reveal the role of LAMP5 in the progression of CRC.Methods GEPIA and LinkedOmics database were used to analyze the expression of LAMP5 in CRC tissues and paired adjacent tissues,and the correlation with clinical parameters and the prognosis of CRC patients followed by validation of clinical samples.Gene Set Enrichment Analysis was used to study the LAMP5 related molecular functions and related signal pathways involved in the occurrence and development in CRC.Results The results of immunohistochemistry showed that the expression of LAMP5 in CRC tissues was significantly higher than that in paired adjacent normal tissues(n=120,P<0.05).LAMP5 expression in CRC tissues was negatively correlated with the degree of differentiation(P<0.05),and positively correlated with N stage and M stage(P<0.05),but not correlated with age,gender and T stage.GEPIA and clinical tissues analysis showed that CRC patients with high expression of LAMP5 had shorter overall survival and disease-free survival than those with low expression of LAMP5(P<0.05).Cox Proportional Hazards Model analysis revealed that the High expression level of LAMP5 was an independent risk factor for OS and DFS in CRC patients(P<0.05).GO analysis showed that LAMP5 had significant correlation with extracellular matrix,extracellular matrix adhesion,extracellular matrix localization,and collagen localization in CRC(P<0.05).KEGG analysis showed that LAMP5 might be involved in multiple pathways,including TGF-βand PI3 K-AKT signaling pathways(P<0.05).Conclusion The expression of LAMP5 is up-regulated in CRC,and correlated with tumor dedifferentiation,lymph node metastasis,distant metastases,and prognosis.Therefore,LAMP5 can be used as a novel biomarker for diagnosis,treatment and prognosis of CRC.
