摘 要:结合超声波提取与纤维素酶法提取紫苏叶多糖,分析酶解时间、酶解温度、加酶量和超声时间等提取条件对紫苏叶多糖提取率的影响,并运用响应面法对紫苏叶多糖提取工艺进行优化。结果表明,当酶解时间为40 min,酶解温度为20.4℃,加酶量为2000 U/g,超声时间为60 min时,紫苏叶多糖的提取率最高,为4.54%。将提取的紫苏叶多糖进行抗氧化活性研究,当紫苏叶多糖溶液质量浓度为0.1 mg/mL时,其DPPH自由基清除率达到94.17%;质量浓度为1 mg/mL时,其羟自由基清除率达到90.06%。This experiment combined ultrasonic-assisted extraction method and cellulase-assisted extraction method to extract polysaccharides from perilla leaves.The response surface analysis was used to optimize the polysaccharide extraction rate with the influence factors such as enzymolysis time,enzymolysis temperature,cellulase quantity and ultrasonic time.The results showsed that the optimum extraction conditions were as follows:enzymolysis time 40 min,enzymolysis temperature 20.4℃,cellulases quantity 2000 U/g and ultrasonic time 60 min.The polysaccharide extraction rate of perilla leaves reacheed 4.54%.The invitro antioxidant activity of the polysaccharides extract indicates when the solution concentration of perilla leaves polysaccharides was 0.1 mg/mL,its DPPH clearance rate reached 94.17%;when the solution concentration was 1 mg/mL,the OH-clearance rate reached 90.06%.
