摘  要:目的观察香附烯酮与卡铂联合应用对三阴性乳腺癌细胞株MDA-MB-231增殖、凋亡的影响,并探讨其机制。方法将MDA-MB-231细胞分为单纯细胞组、卡铂处理组、香附烯酮处理组、香附烯酮联合卡铂组,单纯细胞组加入DMSO,卡铂处理组加入20μmol/L浓度的卡铂,香附烯酮处理组加入0.4 mmol/L浓度的香附烯酮,香附烯酮联合卡铂组加入20μmol/L浓度的卡铂和0.4 mmol/L浓度的香附烯酮。采用CCK-8法测算各组细胞存活率。采用细胞克隆实验观察各组细胞克隆能力,以细胞集落数表示。采用流式细胞术观察各组细胞周期分布情况和细胞凋亡率。采用Western Blotting法检测各组细胞内质网应激相关蛋白PERK、CHOP和凋亡相关蛋白BAX、Bcl-2。结果卡铂处理组、香附烯酮处理组细胞存活率低于单纯细胞组,而香附烯酮联合卡铂组细胞存活率低于其他三组(P均<0.05)。单纯细胞组、卡铂处理组、香附烯酮处理组、香附烯酮联合卡铂组细胞集落数分别为(328±18)、(204±10)、(138±13)、(47±5)个,组间相比,P均<0.05。卡铂处理组将MDA-MB-231细胞周期阻滞在G2/M期,香附烯酮处理组将MDA-MB-231细胞周期阻滞在G0/G1期,香附烯酮联合卡铂组细胞G2/M期和G0/G1期均发生阻滞。卡铂处理组、香附烯酮处理组细胞凋亡率高于单纯细胞组,而香附烯酮联合卡铂组细胞凋亡率高于其他三组(P均<0.05)。与单纯细胞组相比,卡铂处理组、香附烯酮处理组细胞中PERK、CHOP、Bax蛋白相对表达量显著升高,Bcl-2蛋白相对表达量显著降低(P均<0.05);与其他三组相比,香附烯酮联合卡铂组细胞中PERK、CHOP、Bax蛋白相对表达量均显著升高,Bcl-2蛋白相对表达量均显著降低(P均<0.05)。结论香附烯酮与卡铂联合应用可抑制三阴性乳腺癌细胞的增殖,促进其凋亡,其机制可能与调节内质网应激介导的细胞凋亡途径有关。Objective To observe the effects of cyperenone combined with carboplatin on the proliferation and apoptosis of triple-negative breast cancer cell line MDA-MB-231,and to explore their mechanism.Methods MDA-MB-231 cells were divided into four groups:simple cell group,carboplatin-treated group,cyperenone-treated group,and cyperenone combined with carboplatin group.DMSO was added to the simple cell group,carboplatin(20μmol/L)was added to the carboplatin group,cyperenone(0.4 mmol/L)was added to the carboplatin-treated group,and carboplatin(20μmol/L)and cyperenone(0.4 mmol/L)were added to the carboplatin combined with carboplatin group.The cell survival rate of each group was measured by CCK-8 assay.The cell cloning ability of each group was observed by cell cloning experiment,which was expressed by cell colony number.The distribution of cell cycle and the rate of apoptosis were observed by flow cytometry.The endoplasmic reticulum stress-related proteins PERK,CHOP and apoptosis-related proteins BAX and Bcl-2 were detected by Western blotting.Results The cell survival rates of the carboplatin group and cyperenone group were lower than that of the simple cell group,while the cell survival rate of the cyperenone combined with carboplatin group was lower than those of the other three groups(all P<0.05).The number of cell colonies in the simple cell group,carboplatin-treated group,cyperenone-treated group,and cyperenone combined with carboplatin group were 328±18,204±10,138±13 and 47±5,respectively,and there was significant difference between groups(all P<0.05).The cell cycle of MDA-MB-231 was blocked in the G_(2)/M phase in the carboplatin group,MDA-MB-231 cell cycle was blocked in the G_(0)/G_(1) phase in the cyperenone group,and cell cycle of MDA-MB-231 was both blocked in the G_(2)/M phase and G_(0)/G_(1) phase in the cyperenone combined with carboplatin group.The apoptosis rates of the carboplatin group and cyperenone group were higher than that of simple cell group,while the apoptosis rate of the cyperenone comb
