摘 要:目的 探讨NFE2L2对人食管鳞状细胞癌(esophageal squamous cell carcinoma,ESCC)细胞株ECA109侵袭、迁移、增殖、凋亡等生物学行为的影响。方法 基因转染技术下调及上调ECA109细胞中NFE2L2的表达后,Transwell检测细胞侵袭能力及迁移能力,CKK-8检测细胞增殖活性,流式细胞术检测细胞凋亡情况。结果 下调ECA109细胞中NFE2L2的表达后,NFE2L2在siRNA1、siRNA2组基因(0.216±0.084、0.215±0.060)及蛋白(0.238±0.056、0.189±0.023)水平表达降低,细胞侵袭数目[(40.00±8.05)个、(23.00±6.38)个]较对照组[(152.25±27.04)个、(151.00±18.96)个]显著减少,细胞迁移数目[(76.20±20.43)个、(150.33±34.83)个]较对照组[(462.75±59.61)个、(492.40±82.97)个]显著减少,G_(0)/G_(1)期细胞百分比[(68.07±3.45)%、(74.90±4.17)%]较对照组[(54.40±3.91)%、(53.97±2.89)%]明显增加,S期细胞百分比[(30.43±0.91)%、(21.87±4.67)%]较对照组[(42.23±3.12)%、(41.97±3.70)%]明显减少,细胞凋亡率[(4.60±0.61)%、(8.13±0.55)%]较对照组[(2.23±0.74)%、(2.23±0.45)%]明显增加,差异均有统计学意义(P<0.05)。结论 在ECA109细胞中,沉默NFE2L2可抑制其癌细胞的侵袭、迁移及增殖,同时可增加其凋亡,NFE2L2可能在ESCC进展中起促进作用。Objective To investigate the effect of NFE2L2 in the biological behavior (invasion,migration,proliferation,apoptosis,and so on) of esophageal squamous cell carcinoma (ESCC) cell line ECA109.Methods Down-regulating and up-regulating NFE2L2 expression in ECA109 by gene transfection,invasion and migration ability were tested by Transwell assay,proliferation was measured by CKK-8 assay,flow cytometry examined the apoptosis.Results After downregulating NFE2L2 in ECA109,the gene expression of NFE2L2 decreased at siRNA1,siRNA2 (0.216±0.084,0.215±0.060) and protein (0.238±0.056,0.189±0.023),invaded cells (40.00±8.05,23.00±6.38) were diminished than control groups (152.25±27.04,151.00±18.96),migrated cells (76.20±20.43,150.33±34.83) were diminished than control groups (462.75±59.61,492.40±82.97),the cell percent in G_(0)/G_(1)phase[(68.07±3.45)%,(74.90±4.17)%]was increased compared with control groups[(54.40±3.91)%,(53.97±2.89)%].The cell percent in S phase[(30.43±0.91)%,(21.87±4.67)%]was decreased compared with control groups[(42.23±3.12)%,(41.97±3.70)%],the apoptosis rate[(4.60±0.61)%,(8.13±0.55)%]was higher than control groups[(2.23±0.74)%,(2.23±0.45)%](P<0.05).Conclusion Silencing NFE2L2 gene can inhibit the invasion,migration and proliferation of ECA109 cell line,increase the apoptosis of the cell,NFE2L2 gene may promote the progression of ESCC.
