摘 要:目的探讨电针脱敏干预鼻炎大鼠外周和中枢致敏的神经内分泌机制。方法将SD大鼠30只,随机分为正常组(NOR组),模型组(MOD组)和电针组(ACU组),每组10只。MOD组和ACU组分别在第1~10天将卵白蛋白溶液150μl滴入大鼠鼻腔中,NOR组则于鼻腔中滴入等量的生理盐水。采用RT-PCR法监测血清中IL-2及IL-10含量,采用免疫印迹Western blot法测定蛋白表达量。结果在实验造模第10天后,与NOR组和ACU组比较,MOD组的鼻痒、鼻涕、鼻分泌物、行为和总分整体升高(P<0.05)。大鼠抓鼻子次数增多,鼻涕会蔓延至面部,打喷嚏次数增多(P<0.05)。RT-PCR结果显示,MOD组的IL-2和IL-10基因表达量与NOR组和ACU组比较降低(P<0.05);ACU组的IL-2和IL-10基因表达量与MOD组比较恢复明显(P<0.05)。WB结果显示,与NOR组比较,MOD组大鼠脑组织中的EGF和EGFR蛋白表达量升高(P<0.05);ACU组大鼠脑组织中的相应蛋白表达量与MOD组比较降低(P<0.05)。结论电针治疗可脱敏干预鼻腔变应性鼻炎,从外周及中枢影响免疫因子及蛋白分泌。Objective To investigate the neuroendocrine mechanism by which electroacupuncture desensitizationintervenes in peripheral and central sensitization of rhinitis rats.Methods Thirty SD rats were randomly divided into the NOR group,MOD group and ACU group,with 10 rats in each.In the MOD group and ACU group,150μl ovalbuminsolution was dripped into the nasal cavity of rats from day 1 to 10,respectively.The NOR group received nasal drip ofnormal saline ofthe same amount.The contents of IL-2 and IL-10 in serum were detected by RT-PCR,and the proteinexpression by Western blot.Results Ten days into the modeling,the overall increase in nasal itching,nasal discharge,behavior and total scores was observed in the MOD group compared with the NOR group and ACU group(P<0.05).When noses were scratchedmore often,nasal mucus spread across the face and the number of times of sneezing increased(P<0.05).Rt-PCR results showed that IL-2 and IL-10 gene expressionsin the MOD group were lower than those of the NOR group and ACU group(P<0.05),but they were significantly recovered in the ACU group compared with the MOD group(P<0.05).Western blot results showed that compared with the NOR group,EGF and EGFR protein expressions in the MOD group were increased(P<0.05).The expressions of corresponding proteins in the ACU group were lower than in the MOD group(P<0.05).Conclusion Electroacupuncture treatment canintervenein nasal allergic rhinitis via desensitization and affect immune factors and protein secretion from peripheral and central systems.
