摘 要:目的研究阿尔茨海默病大鼠肠道菌群的变化,观察菌群组成变化。方法将30只大小、年龄、体质量相似的大鼠应用阿尔茨海默病造模方法,并且在造模内28 d监测其体质量、血糖、血压的变化,对变化不稳定的大鼠进行淘汰,应用Morris水迷宫测试方法将其分为3组,分别为对照组、轻症模型组、重症模型组,每组6只。提取对照组、轻症模型组和重症模型组大鼠的粪便样本,进行16S rRNA测序检测,并进行物种组成分析、Alpha多样性分析、Beta多样性分析和关联网络分析。结果在门水平上模型组中硬壁菌门(Firmicutes)、螺旋体门(Spirochetes)丰度降低,而拟杆菌门(Bacteroidetes)丰度上升,在属水平上以普雷沃氏菌属(Prevotella)和乳杆菌属(Lactobacillus)变化最为明显。阿尔茨海默病大鼠的肠道菌群多样性和丰富度明显改变。结论重症模型组与轻症模型组肠道菌群发生改变,包括多样性和丰富度组成变化,且轻症模型组与重症模型组之间也存在差异。Objective To study the changes of intestinal microbiota in Alzheimer’s disease rats and observe the changes of bacterial composition.Methods A total of 30 rats with similar size,age,weight were modeled with Alzheimer’s disease and the changes of weight,blood glucose and blood pressure were monitored for 28 days.The unstable rats were eliminated.The rest rats were divided into three groups by Morris water maze test,control,mild model group and severe model group,with six rats in each group.Fecal samples from rats with control,mild model group and severe model group were extracted for 16S rRNA sequencing,and species composition analysis,Alpha diversity analysis,Beta diversity analysis and association network analysis.Results The results showed that the Firmicutes and Spirochetes abundance decreased,and the abundance of Bacteroidetes increased in the model group at the phylum level,with the Prevotella and Lactobacillus changes being the most obvious at the genus level.The intestinal microflora diversity and richness were altered in Alzheimer’s disease rats.Conclusion The results showed that the intestinal microflora was altered between the severe model group and mild model group,including diversity and richness composition changes,and that there were also differences between mild model group and severe model group.
