摘 要:目的 探讨慢性牙周炎(CP)妊娠大鼠胎盘中Toll样受体4(TLR4)/核因子-κB(NF-κB)通路对炎症反应的调控作用。方法 将雌性SD大鼠随机分为对照组、CP组、DMSO对照组、CP+DMSO组、CP+TAK242组,对照组、DMSO对照组进行假手术操作,CP组、CP+DMSO组、CP+TAK242组进行CP造模,4周后与雄性SD大鼠合笼,妊娠后DMSO对照组及CP+DMSO组给予对照溶剂DMSO腹腔注射、CP+TAK242组给予TLR4拮抗剂TAK242腹腔注射。观察早产率,称量新生鼠体质量,检测胎盘中TLR4、NF-κB、Myd88的表达水平,血清及胎盘中TNF-α、IL-6的含量。结果 CP组大鼠的早产率87.5%、高于对照组的12.5%,胎盘中TLR4、NF-κB、Myd88的表达水平,血清及胎盘中TNF-α、IL-6的含量均明显高于对照组;新生鼠体质量为(5.09±0.84)g,明显低于对照组的(7.51±1.03)g(P<0.05)。CP+TAK242组大鼠的早产率为37.5%,低于CP+DMSO组的87.5%;胎盘中NF-κB、Myd88的表达水平,血清及胎盘中TNF-α、IL-6的含量均明显低于CP+DMSO组;新生鼠体质量为(7.09±1.32)g,明显高于CP+DMSO组的(5.29±0.88)g(P<0.05)。结论 CP妊娠大鼠胎盘中TLR4/NF-κB通路激活对胎盘及全身炎症反应的激活促进作用,进而可能参与早产的发生。Objective To study the regulation effect of toll like receptor 4(TLR4)/nuclear factor-κB(NF-κB pathway in placenta of pregnant rats with chronic periodontitis(CP)on inflammatory response. Methods Female SD rats were randomly divided into control group,CP Group,DMSO control group,CP+DMSO group and CP+TAK242 group. Sham operation was performed in the control group and DMSO control group. CP model was made in CP Group,CP+DMSO group and CP+TAK242 group. After 4 weeks,they were caged with male SD rats. After pregnancy,DMSO control group and CP+DMSO group were injected intraperitoneally with control solvent DMSO CP+TAK242 group was injected intraperitoneally with TLR4 antagonist TAK242. The premature birth rate was observed,the body mass of neonatal rats was weighed,and the expression of TLR4,MyD88 and NF-κB,the contents of TNF-α,IL-6 in serum and placenta were detected. Results The premature birth rate 87.5% of CP group were higher than 12.5% of control group,the expression of TLR4,MyD88 and NF-κB,the contents of TNF-α,IL-6 in serum and placenta of CP group were higher than those of control group,the body mass of neonatal rats(5.09±0.84)g was lower than(7.51±1.03)g of control group(P<0.05). The premature birth rate 37.5% of CP+TAK242 group was lower than 87.5% of CP+DMSO group,the expression of TLR4,MyD88 and NF-κB,the contents of TNF-α,IL-6 in serum and placenta of CP+TAK242 group were lower than those of CP+DMSO group,the body mass of neonatal rats(7.09±1.32)g was higher than(5.29±0.88)g of CP+DMSO group(P<0.05). Conclusion The activation of TLR4/NF-κB pathway in placenta of CP pregnant rats promotes placental and systemic inflammatory response,which may be involved in the occurrence of preterm birth.
