摘  要:目的研究卵巢肿瘤中自噬相关基因7(ATG7)的表达情况,并分析ATG7对卵巢癌细胞SKOV3增殖的影响及相关机制。方法收集2022年2月至4月蚌埠医学院第一附属医院肿瘤妇科手术切除卵巢癌组织及癌旁正常组织15例,实时荧光定量PCR(qRT-PCR)检测卵巢癌及癌旁正常组织中ATG7表达差异,siRNA转染卵巢癌细胞SKOV3,敲低ATG7表达量,Western blot验证转染效率。通过克隆形成实验检测SKOV3细胞增殖变化。采用Western blot检测磷酸化磷脂酰肌醇3-激酶(PI3K)/蛋白激酶B(AKT)相关信号通路的蛋白表达变化。结果与癌旁正常组织相比,卵巢癌组织中ATG7蛋白表达增高(P<0.05),敲低SKOV3细胞ATG7表达量,细胞增殖能力降低(P<0.05),且敲低后的p-PI3K和p-AKT蛋白表达量下调(P<0.05)。结论ATG7蛋白表达在卵巢癌中明显升高,且可能通过PI3K/AKT信号通路影响卵巢癌细胞SKOV3增殖。Objective To study the expression of autophagy-related gene(ATG7)in ovarian tumors,and to analyze the effect of ATG7 on SKOV3 proliferation and its related mechanism.Methods From February to April 2022,15 cases of ovarian cancer tiss ue and adjacent normal tiss ue were collected from the Department of Gynecological Oncology of the First Affiliated Hospital of Bengbu Medical College,and real-time fluorescent quantitative PCR(qRT-PCR)was used to detect the expression differences of ATG7 in ovarian cancer and adjacent normal tissue.SKOV3 was transfected with siRNA,and the expression level of ATG7 was knocked down.Western blot was used to verify the transfection efficiency.The proliferation of SKOV3 cells was detected by clonal formation assay.Western blot was used to detect the protein expression of phosphoinositide 3-kinase(PI3K)/protein kinase B(AKT)related signaling pathway.Results Compared with normal adjacent tissues,the expression of ATG7 protein in ovarian cancer tissues increased(P<0.05),and the cell proliferation ability of SKOV3 cells decreased by knocking down the expre ssion of ATG7 protein(P<0.05),and the protein expressions of p-PI3K and p-AKT were down-regulated after knocking down(P<0.05).Conclusion The expression of ATG7 protein is significantly increased in ovarian cancer and may affect SKOV3 proliferation through PI3K/AKT signaling pathway.
