肝癌介入治疗患者组织miR-424表达水平及与肿瘤复发的关系分析

摘　　要：目的分析肝癌介入治疗患者组织miR-424表达水平及与肿瘤复发的关系。方法选取新乡医学院第三附属医院2017年10月—2020年10月60例肝癌行介入治疗患者作为观察对象。收集患者临床资料。实时荧光定量检测患者肝组织中miR-424表达水平。对患者进行为期1年的随访,分析miR-424表达水平与治疗后复发及生存的关系。蛋白免疫印迹检测肝组织生长抑制因子4(ING4)蛋白相对表达水平。结果肝癌患者治疗后miR-424 mRNA和ING4蛋白相对表达水平明显高于治疗前(P<0.05)。miR-424低表达患者和miR-424高表达患者在年龄、性别、肝功能Child pugh分级、微血管侵犯和血清甲胎蛋白(AFP)水平比较,差异无统计学意义(P>0.05)。miR-424低表达患者和miR-424高表达患者在肿瘤分化程度、TNM分级和肿瘤直径方面比较,差异具有统计学意义(P<0.05)。miR-424低表达组患者治疗后6个月、治疗后1年复发率明显高于miR-424高表达组(P<0.05)。随访1年,死亡19例,存活41例;死亡组患者肝癌组织miR-424 mRNA和ING4蛋白相对明显低于存活组(P<0.05)。结论肝脏组织miR-424表达水平是肝癌患者复发的风险因素,同时可作为预后的诊断指标。Objective To analyze the expression level of miR-424 in tissues and its relationship with tumors recurrence in patients with liver cancer undergoing intervention therapy.Methods A total of 60 patients with liver cancer undergoing intervention therapy in the hospital were enrolled as observation group between October 2017 and October 2020,while other 30 patients with hepatitis and cirrhosis during the same periodwere enrolled as control group.The clinical data of patients were collected.The expression level of miR-424 inliver tissues was detected by real-time fluorescence quantitative method.The patients were followed up for 1 year to analyze the relationship between miR-424 level and postoperative recurrence,survival.The relative expression level of inhibitor of growth 4(ING4)protein was detected by Western blot.Results The relative expression levels of miR-424 mRNA and ING4 protein in patients with liver cancer after treatment were significantly higher than those before treatment(P<0.05).There was no significant difference in age,gender,Childpugh grading of liver function,microvascular invasion or level of serum AFP between patients with low-expression miR-424 and those with high-expression miR-424(P>0.05),but differences in tumor differentiation,TNM grading and tumor diameter were statistically significant(P<0.05).The recurrence rates in miR-424 low-expression group were significantly higher than those in miR-424 high-expression group at 6 months and 1 year after surgery(P<0.05).After 1 year of follow-up,there were 19 died cases and 41 survivedcases.The relative expression levels of miR-424 mRNA and ING4 protein in death group were significantly lower than those in survival group(P<0.05).Conclusion The expression level of miR-424 in liver tissues is a risk factor of recurrence in liver cancer patients,which can be applied as a diagnostic index for prognosis.
