摘  要:运用扫描电镜观察了五种不同成分体系高含水量水凝胶样品的三维结构,结果表明:风干法、烘干法和冷冻干燥法不适合用于水凝胶扫描电镜样品的制备;包埋有活细胞的水凝胶样品,用CO临界点干燥法制备,细胞形态良好,但水凝胶三维结构坍塌、失真;运用薄层制样技术,无须添加附属设备,即可得到保存精良的水凝胶三维结构和细胞形貌,实现对水凝胶样品的分析测试。该方法能真实反映水凝胶的形貌,有效解决了对高含水样品三维结构研究的技术难题。The three-dimensional structures of five hydrogels with high water content were observed by scanning electron microscope. The results showed that air-drying, lamp drying and freeze-drying methods are not suitable for the preparation of hydrogel samples for scanning electron microscope. If a hydrogel sample containing living cells, prepared by COcritical point drying method, was observed under scanning electron microscope, the morphology of the cells was good, but the three-dimensional structure of hydrogel collapsed and distorted. With thin-layer sample preparation technology and no additional equipment, well-preserved hydrogels and cell morphology can be obtained to realize the analysis and the test of hydrogels samples. This method can truly reflect the morphology of hydrogel and effectively solve the technical difficulties in the study of three-dimensional structure of hydrogels with extremely high water content.
