摘 要:以丁二酸(SA)和4-氨基安替吡啉(AAP)为配体构筑Co(Ⅱ)配合物,以鲱鱼精DNA为靶点,通过紫外吸收光谱、循环伏安法和DNA粘度滴定实验,探讨了配合物与鲱鱼精DNA之间相互作用的机制,并采用抑菌圈法测试了配合物对金黄色葡萄球菌(St.aureus)的抑菌性能。结果表明:配合物中加入DNA后,吸收峰出现明显的减色和红移,减色和红移程度与DNA加入量呈正相关;配合物的氧化还原峰的峰电位发生正移,峰电流减小;DNA的粘度随配合物加入量的增大而增大。因此,可推断配合物以嵌插方式与DNA形成1∶1的复合物,其结合常数为3.77×10^(3) L·mol^(-1)。配合物对St.aureus具有显著的抑菌效果。A kinds of cobalt complex of succinic acid and 4-aminoanantipyridinyl was synthesized firstly.In addition,with herring sperm DNA as the research target,the interaction with DNA was investigated by UV absorption spectroscopy,CV method and DNA viscosity titration experiment,which provided the basis for the design and development of new anticancer drugs.Results showed that the UV absorption peaks of complex addition DNA was red-shifted and the intensity was weakened.The peak current decreased significantly and potential moved positively,and the relative viscosity of DNA was increased.All the above indicated that the interaction mode of the complex with DNA belonged to intercalative,the combining ratio and the binding constant was 1∶1 and 1.55×10^(4) L·mol^(-1),respectively.Moreover,the complexes had significant antibacterial effects on St.Aureus.
