摘 要:为了解临床多重耐药铜绿假单胞菌的全基因组特征并探讨其多重耐药机制,利用Illumina PE测序平台对1株来自烧伤病人下肢分泌物中的多重耐药铜绿假单胞菌PA803进行全基因组测序,并通过生物信息学方法对分离菌株进行群体进化和分型分析以及基因功能注释。结果发现,PA803基因组大小为7040317 bp,GC含量为66.8%,预测编码6528个基因,包含60个tRNA、2个rRNA。COG比对基因对应许多未知功能蛋白,KEGG分析发现与细菌耐药相关的生物膜形成、双组分系统和外排泵等代谢途径较为活跃,与数据库比对其序列分型为ST620,共有90个耐药基因和357个毒力因子基因,并构建系统进化树,显示其与美国2株铜绿假单胞菌菌株亲缘关系较近。临床多重耐药菌株给患者治愈造成很大困难,涉及多个基因及多种耐药机制,各机制之间的关系还需进一步研究。To understand the whole genome characteristics of clinical multi drug resistant Pseudomonas aeruginosa and explore its multi drug resistance mechanism,the whole genome of a multi drug resistant Pseudomonas aeruginosa PA803 strain from the lower limb secretion of burn patients was sequenced by Illumina PE sequencing platform.The population evolution,typing and gene function annotation of the isolated strains were analyzed by bioinformatics methods.The genome size of PA803 was 7040317 bp and the GC content was 66.8%.It was predicted to encode 6528 genes,including 60 tRNAs and 2 rRNAs.COG alignment genes correspond to many unknown functional proteins.KEGG analysis found that the metabolic pathways related to bacterial drug resistance,such as biofilm formation,two-component system and efflux pump,were more active.Compared with the database,its sequence typing was ST620,with a total of 90 drug resistance genes and 357 virulence factor genes.The phylogenetic tree was constructed,which showed that it was closely related to two strains of Pseudomonas aeruginosa in the United States.Clinical multi drug resistant strains cause great difficulties for patients to cure,involving multiple genes and multiple drug resistance mechanisms,and the relationship between the mechanisms needs to be further studied.
