摘 要:目的探讨TSPAN13对前列腺癌细胞增殖、迁移和侵袭的影响。方法将PC3细胞、22Rv1细胞、LNCAP细胞、DU145细胞用含10%胎牛血清和1%青链霉素双抗的RPMI1640培养基培养,BPH-1细胞用含10%胎牛血清和1%青链霉素双抗的DMEM培养基培养,培养环境均为37℃、5%CO_(2)、饱和湿度。采用RT-PCR和Westernblotting实验检测TSPAN13表达;通过si-RNA沉默TSPAN13检测TSPAN13功能;通过CCK-8实验和集落形成实验检测细胞的增殖和克隆形成能力;通过划痕实验和Transwell实验检测细胞的迁移和侵袭能力。结果TSPAN13蛋白在前列腺癌中高表达(P<0.01);TSPAN13 mRNA和蛋白在PC3和DU145细胞中的表达高于BPH-1细胞(P均<0.01);沉默TSPAN13,PC3和DU145细胞中TSPAN13 mRNA和蛋白表达降低;CCK-8和集落形成实验显示,PC3和DU145细胞增殖能力和克隆生成能力下降(P均<0.01);划痕实验和Transwell实验显示,PC3和DU145细胞的迁移和侵袭能力均降低(P均<0.01)。结论沉默TSPAN13表达可抑制前列腺癌PC3和DU145细胞增殖、克隆形成、迁移、侵袭能力,TSPAN13可能在前列腺癌发生、发展中发挥重要作用。Objective To explore the effects of TSPAN13 on the proliferation,migration,and invasion of prostate cancer cells.Methods PC3 cells,22Rv1 cells,LNCAP cells,and DU 145 cells were cultured in RPMI 1640 medium containing 10%fetal bovine serum and 1%penicillin-streptomycin double antibody.BPH-1 cells were cultured in DMEM medium containing 10%fetal bovine serum and 1%penicillin-streptomycin double antibody.The culture temperature was 37℃,the carbon dioxide concentration was 5%and the humidity was saturated.The expression of TSPAN13 was detected by RT-PCR and Western blotting;the function of TSPAN13 was detected by silencing TSPAN13 with si-RNA;the prolifer⁃ation and clone formation of cells were detected by CCK-8 and clone formation assay;the migration and invasion of cells were detected by Scratch test and Transwell test.Results TSPAN13 protein was highly expressed in prostate cancer(P<0.01);the expression levels of TSPAN13 mRNA and protein were significantly higher in the PC3 and DU145 cells than in BPH-1 cells(all P<0.01).After TSPAN13 was silenced,the mRNA and protein expression levels of TSPAN13 decreased in PC3 and DU145 cells.CCK-8 and clone formation assay showed that the proliferation and clone formation of PC3 and DU145 cells decreased(both P<0.01).Scratch test and Transwell test showed that the migration and invasion of PC3 and DU145 cells decreased(both P<0.01).Conclusion Silencing TSPAN13 inhibits the proliferation,clone formation,migration and invasion of PC3 and DU145 cells,and TSPAN13 may play an important role in the development and progression of prostate cancer.
