摘　　要：目的探讨环状RNA(circRNA)hsa_circ_0004771基因干扰对骨肉瘤MG63细胞增殖和凋亡的影响。方法用hsa_circ_0004771基因干扰序列(shRNA)及阴性对照序列(shRNA-NC)分别转染MG63细胞,同时设空白对照组(不转染)。EdU染色法检测细胞增殖情况;流式细胞术及Hoechst染色法检测细胞凋亡情况;流式分选法检测线粒体膜电位;相应试剂盒检测丙二醛(malondialdehyde,MDA)及乳酸脱氢酶(lactate dehydrogenase,LDH)含量;Western blot法检测Bax、Bcl-2、Caspase-3、cleaved Caspase-3、c-Myc、蛋白表达水平。结果与空白对照组比较,shRNA组EdU阳性细胞数明显降低(P<0.05),细胞凋亡率明显升高(P<0.05),线粒体膜电位明显降低(P<0.05),MDA及LDH含量显著升高(P均<0.05),Bax/Bcl-2及cleaved Caspase-3/Caspase-3比值明显升高(P均<0.05),c-Myc蛋白表达水平显著降低(P<0.05)。结论circRNA hsa_circ_0004771基因干扰可抑制骨肉瘤细胞MG63增殖,并诱导细胞凋亡,降低线粒体膜电位。Objective To investigate the effect of circRNA hsa_circ_0004771 gene interference on the proliferation and apoptosis of osteosarcoma MG63 cells.Methods MG63 cells were transfected with hsa_circ_0004771 gene interference sequence(shRNA)and negative control sequence(shRNA-NC)respectively,using those untransfected as blank control.Cell proliferation was determined by EdU staining,while cell apoptosis by flow cytometry and Hoechst staining,mitochondrial membrane potential by flow separation,the contents of MDA and LDH by the corresponding kits,and the expression levels of Bax,Bcl-2,Caspase-3,cleaved Caspase-3 and c-Myc proteins by Western blot.Results Compared with those in blank control group,the count of EdU positive cells in shRNA group decreased significantly(P<0.05),while apoptosis rate increased significantly(P<0.05),mitochondrial membrane potential decreased significantly(P<0.05),the contents of MDA and LDH increased significantly(each P<0.05),the ratios of Bax to Bcl-2 and cleaved Caspase-3 to Caspase-3 increased significantly(each P<0.05).However,the expression level of c-Myc protein decreased significantly(P<0.05).Conclusion The circRNA hsa_circ_0004771 gene interference inhibits the proliferation,induces the apoptosis and decreases the mitochondrial membrane potential of osteosarcoma MG63 cells.
