摘  要:采用牛乳酪蛋白糖巨肽制备酪蛋白糖肽,并分析其对二肽基肽酶-Ⅳ(dipeptidyl peptidase-Ⅳ, DPP-Ⅳ)活性的抑制作用,筛选DPP-Ⅳ抑制活性最强的酪蛋白糖肽,并进行分离鉴定。结果表明:(1)酪蛋白糖肽的DPP-Ⅳ抑制率最高达54.55±3.65%;(2)经葡聚糖凝胶G-25(Sephadex G-25)分离得到两个组分,其中F1组分具有较强的DPP-Ⅳ抑制活性,抑制率达63.27±3.24%;(3)进一步利用反相高效液相色谱及液相色谱串联质谱从F1组分中分离并鉴定得到3条DPP-Ⅳ抑制肽,肽序列分别为MAIPPKKNQDKT、PPKKNQDKTEIPTI、SPEVIESPPEIN。研究结果表明酪蛋白糖巨肽经酶解获得的酪蛋白糖肽具有DPP-Ⅳ抑制活性,具备作为降糖活性功能性配料开发的潜在性。The purpose of this study is to investigate the inhibitory effect of casein glycopeptide on the activity of dipeptidyl peptidase-Ⅳ(DPP-Ⅳ). Casein glycopeptide was prepared and the highest DPP-Ⅳ inhibitory activity peptides were screened for isolation and identification. The results showed that casein glycopeptide exhibited the greatest DPP-Ⅳ inhibitory activity(54.55 ±3.65%). Casein glycopeptide were separated into two fractions by Sephadex G-25 gel filtration, among which fraction F1 showed a stronger DPP-Ⅳ inhibitory activity(63.27±3.24%). Then three peptides with potential DPP-Ⅳ inhibitory activity were isolated and identified by reversedphase high performance liquid chromatography(RP-HPLC) and liquid chromatography electron spray ionization mass/mass(LC-ESI-MS/MS). The peptide sequences were MAIPPKKNQDKT, PPKKNQDKTEIPTI and SPEVIESPPEIN, respectively. This study proved that casein glycopeptide showed DPP-Ⅳ inhibitory activity and may possess potential as functional dairy ingredients.
