摘  要:目的研究冬凌草甲素通过circ-LRBA调控miR-4735-3p对膀胱癌细胞生物学功能和糖酵解影响。方法实时PCR方法检测膀胱癌组织和细胞中circ-LRBA表达变化。膀胱癌细胞SCaBER分成对照组、si-NC组、si-circ-LRBA组、ORI组、ORI+vector组、ORI+circ-LRBA组、ORI+anti-NC组、ORI+anti-miR-4735-3p组、ORI+circ-LRBA+miR-NC组和ORI+circ-LRBA+miR-4735-3p组。CCK-8实验检测细胞增殖,流式细胞术检测细胞凋亡,用试剂盒检测细胞ATP水平、乳酸产量和葡萄糖消耗量。荧光素酶报告系统鉴定circ-LRBA和miR-4735-3p的靶向关系。结果膀胱癌组织和细胞中circ-LRBA表达水平升高(P<0.05)。与对照组比较,si-circ-LRBA组和ORI组膀胱癌细胞SCaBER中circ-LRBA表达水平降低,细胞存活率下降,细胞凋亡率升高,ATP水平降低,乳酸生成量减少,葡萄糖消耗量降低(P<0.05)。与ORI+vector组比较,ORI+circ-LRBA组膀胱癌细胞SCaBER中circ-LRBA表达水平升高,细胞存活率升高,凋亡率降低,ATP水平升高,乳酸生成量和葡萄糖消耗量升高(P<0.05);与ORI+anti-NC组比较,ORI+anti-miR-4735-3p组膀胱癌细胞SCaBER中miR-4735-3p表达水平降低,细胞存活率升高,凋亡率降低,ATP水平升高,乳酸生成量和葡萄糖消耗量升高(P<0.05);与ORI+circ-LRBA+miR-NC组比较,ORI+circ-LRBA+miR-4735-3p组膀胱癌细胞SCaBER中miR-4735-3p表达水平升高,细胞存活率降低,凋亡率升高,ATP水平降低,乳酸生成量和葡萄糖消耗量降低(P<0.05)。Circ-LRBA靶向负调控miR-4735-3p。结论冬凌草甲素通过下调circ-LRBA促进miR-4735-3p表达诱导膀胱癌细胞SCaBER凋亡,抑制细胞增殖和糖酵解。Objective To study the effects of oridonin regulating miR-4735-3 p through circ-LRBA on the biological function and glycolysis of bladder cancer.Methods Real-time PCR method was used to detect the expression changes of circ-LRBA in bladder cancer tissues and cells.Bladder cancer cells SCaBER was divided into control group,si-NC group,si-circ-LRBA group,ORI group,ORI+vector group,ORI+circ-LRBA group,ORI+anti-NC group,ORI+anti-miR-4735-3 p group,ORI+circ-LRBA+miR-NC group,ORI+circ-LRBA+miR-4735-3 p group.CCK-8 assay was used to detect cell proliferation,and the flow cytometry was used to detect cell apoptosis.the levels of ATP,lactate production and glucose consumption were detected by kits.The luciferase reporter system was uesd to identify the targeting relationship between circ-LRBA and miR-4735-3 p.Results The expression levels of circ-LRBA in bladder cancer tissues and cells were increased(P<0.05).Compared with the control group,the expression levels of circ-LRBA in si-circ-LRBA group and ORI group were decreased,cell survival rates were decreased,cell apoptosis rates were increased,ATP levels were decreased,lactate productions were decreased,glucose consumptions were decreased(P<0.05).Compared with the ORI+vector group,the expression level of circ-LRBA in ORI+circ-LRBA group was increased,cell survival rate was increased,cell apoptosis rate was decreased,ATP level was increased,lactate production was increased,glucose consumption was increased(P<0.05).Compared with the ORI+anti-NC group,the expression level of miR-4735-3 p in bladder cancer cell SCaBER in the ORI+anti-miR-4735-3 p group was decreased,cell survival rate was increased,apoptosis rate was decreased,ATP level was increased,and lactate production and glucose consumption was increased(P<0.05);compared with the ORI+circ-LRBA+miR-NC group,the expression level of miR-4735-3 p in bladder cancer cell SCaBER was increased in the ORI+circ-LRBA+miR-NC group,the survival rate was decreased,the apoptosis rate was increased,the ATP level was decreased,the prod
