摘 要:红托竹荪菌托多糖具有明显的生物活性,其提取方法常涉及浓缩及醇沉步骤。为提高红托竹荪菌托多糖的得率,文章采用单因素试验结合正交试验设计优化红托竹荪菌托多糖的浓缩及醇沉工艺。结果显示:浓缩比(原料质量∶浓缩液体积,g/mL)为1∶2时,多糖得率最高,正交试验优化的最佳醇沉条件为:醇沉时间2 h、乙醇浓度85%、4倍体积乙醇用量,此条件下,所得多糖质量最大。该研究可为红托竹荪菌托多糖精深加工中浓缩及醇沉工艺提供理论指导,促进红托竹荪菌托的废物利用。Polysaccharides from Dictyophora rubrovalvata volva have obvious biological activity,and the extraction methods often involve the steps of concentration and alcohol precipitation.In order to improve the yield of polysaccharides,single factor experiment combined with orthogonal experiment design is used to optimize the concentration and alcohol precipitation process of polysaccharides.The results show that the yield of polysaccharides is the highest when the concentration ratio(raw material mass∶concentrated solution volume,g/mL)is 1∶2.The optimal conditions of alcohol precipitation by orthogonal experiment optimization are as follows:alcohol precipitation time is 2 h,ethanol concentration is 85%,ethanol dosage is 4 times of volume.Under such conditions,the mass of polysaccharides is the highest.This research can provide theoretical guidance for the concentration and alcohol precipitation process of polysaccharides from Dictyophora rubrovalvata volva in the intensive processing,and promote the waste utilization of Dictyophora rubrovalvata volva.
