摘 要:目的通过生物信息学的方法以及临床标本回顾性分析研究了肝配蛋白A2(EFNA2)基因在肺腺癌中表达与免疫浸润的临床意义。方法利用仙桃生物信息学工具分析EFNA2在LUAD中的表达。使用Cox回归分析与TCGA患者总生存期相关的临床病理学特征。采用Log Rank分析研究数据库及临床标本不同EFNA2表达水平患者的OS。使用ssGSEA算法研究EFNA2表达与各种浸润性免疫细胞丰度之间的相关性,了介EFNA2在LUAD中与癌症免疫浸润之间的相关性。结果基于从TCGA和GTEx获得的数据的非配对及配对分析结果证实,EFNA2在LUAD中的表达显着上调。Logistic分析发现pT分期(P=0.010)是EFNA2表达的最终影响因素。Log-Rank检验结果表明LUAD中EFNA2的低表达与良好的预后相关。cox回归分析显示pT分期(P<0.001)、pN分期(P<0.001)和EFNA2表达(P=0.030)是OS的最终影响因素。ssGSEA算法分析免疫浸润细胞的丰度结果显示,在EFNA2低表达组中T细胞(P<0.05)、肥大细胞(P<0.05)、中性粒细胞(P<0.01)、毒性细胞(P<0.05)和B细胞(P<0.01)的肿瘤浸润丰度明显升高。结论EFNA2可以作为评估LUAD预后和免疫浸润水平的潜在生物标志物。Objective To study the clinical significance of ephrin A2(EFNA2)gene expression and immune infiltration in lung adenocarcinoma(LUAD)by bioinformatics methods and retrospective analysis of clinical specimens.Methods The expression of EFNA2 in LUAD was analyzed using Xiantao bioinformatics tool.Clinicopathological characteristics associated with overall survival(OS)in TCGA patients were analyzed using Cox regression.Log Rank was used to analyze the OS of patients with different EFNA2 expression levels in the research database and clinical specimens.The correlation between EFNA2 in LUAD and cancer immune infiltration was investigated using the ssGSEA algorithm to investigate the correlation between EFNA2 expression and the abundance of various infiltrating immune cells.Results The results of unpaired and paired analysis based on data obtained from TCGA and GTEx confirmed that the expression of EFNA2 was significantly up-regulated in LUAD.Logistic analysis found that pT stage(P=0.010)was the ultimate influencing factor for EFNA2 expression.Log-Rank test results indicated that low expression of EFNA2 in LUAD was associated with good prognosis.Cox regression analysis showed that pT stage(P<0.001),pN stage(P<0.001)and EFNA2 expression(P=0.030)were the final influencing factors of OS.The abundance of immune infiltrating cells analyzed by ssGSEA algorithm showed that T cells(P<0.05),mast cells(P<0.05),neutrophils(P<0.01),and toxic cells(P<0.05)in the low expression group of EFNA2 and B cells(P<0.01)were significantly higher in tumor-infiltrating abundance.Conclusions EFNA2 may serve as a potential biomarker to assess the prognosis and immune infiltration level of LUAD.
