摘  要:目的探究miR-146a/肿瘤坏死因子-α受体相关因子6(TRAF6)信号通路在非小细胞肺癌(NSCLC)组织中的表达及临床病理意义。方法选取2009—2017年手术切除的53例非癌肺组织(Non-CLT)和118例NSCLC组织(其中腺癌54例、鳞癌64例)并回顾性分析其临床病理资料。采用实时荧光定量PCR法和免疫组织化学染色法(IHC)检测组织miR-146a、TRAF6表达,并进一步分析其与临床病理特征、生存预后的相关性。结果与Non-CLT组织相比,NSCLC组织中miR-146a相对表达量降低(P<0.001),TRAF6蛋白阳性表达率升高(P<0.001)。经Spearman秩相关性分析,NSCLC组织中miR-146a相对表达量与TRAF6蛋白IHC评分呈负相关性(r_(s)=-0.515,P<0.001)。TNM分期Ⅲ期及伴有淋巴结转移患者miR-146a低表达率及TRAF6阳性表达率分别高于TNM分期Ⅰ—Ⅱ期、无淋巴结转移者(P<0.001)。miR-146a低表达、TRAF6阳性表达单独或联合表达模式的生存率低于其他表达模式的患者(P<0.05)。miR-146a低表达(P=0.012)和TRAF6阳性表达(P=0.017)的肺腺癌患者的总生存率较低;TRAF6阳性表达与肺鳞癌患者总生存预后不良有关(P=0.007)。在联合情况下,miR-146a低表达/TRAF6阳性表达的肺腺癌和肺鳞癌患者总生存率低于其他患者(P<0.05)。组织TRAF6阳性表达和淋巴结转移是肺鳞癌患者的独立预后因素(P<0.05)。结论NSCLC组织中miR-146a表达降低,TRAF6蛋白表达升高。miR-146a/TRAF6通路可能在NSCLC的发展中发挥着重要作用。Objective To investigate the expression and clinicopathological significance of miR-146a/tumor necrosis factor receptor-associated factor 6(TRAF6)signaling pathway in non-small cell lung cancer(NSCLC).Methods Fifty-three non-cancerous lung tissues(Non-CLT)and 118 NSCLC tissues(54 cases of adenocarcinoma and 64 of squamous cell carcinoma)resected from our hospital from 2009 to 2017 were selected in this study.The clinicopathological data were analyzed retrospectively,and the levels of miR-146a and TRAF6 protein expression were detected by real-time fluorescence quantitative PCR and immunohistochemistry(IHC),respectively.Moreover,the relationships of miR-146a and TRAF6 protein expression to clinicopathological features and survival prognosis were analyzed.Results Compared with non-CLT tissues,miR-146a expression decreased but TRAF6 protein expression increased in NSCLC tissues(P<0.001).Spearman rank correlation analysis showed that miR-146a expression was negatively correlated with IHC score of TRAF6 protein in NSCLC tissues(r_(s)=-0.515,P<0.001).The low miR-146a expression rate and the positive TRAF6 expression rate in patients with stageⅢcancer and lymph node metastasis were higher than those in patients with stageⅠorⅡcancer and without lymph node metastasis(P<0.001).The survival rates of patients with low miR-146a expression,positive TRAF6 expression or their combination were lower than those of patients with other expression patterns(P<0.05).The overall survival rate was low in patients with low miR-146a expression(P=0.012)or positive TRAF6 expression(P=0.017).Positive TRAF6 expression was associated with poor overall survival prognosis in patients with lung squamous cell carcinoma(P=0.007).In addition,the overall survival rate in patients with lung adenocarcinoma and squamous cell carcinoma with low miR-146a expression and positive TRAF6 expression was lower than that in other patients(P<0.05).Positive TRAF6 protein expression and lymph node metastasis were independent prognostic factors for lung squamou
