千金子提取物调控AK125512对肝癌细胞增殖、迁移、侵袭的影响研究

摘　　要：目的 研究千金子提取物在肝癌MHCC97-H细胞增殖、迁移、侵袭和凋亡中的作用与潜在分子机制。方法 用千金子提取物5 mg/mL、10 mg/mL、20 mg/mL分别处理肝癌细胞MHCC97-H,CCK-8法和流式细胞术检测MHCC97-H细胞增殖和凋亡,Transwell实验检测细胞迁移和侵袭能力,Western blot检测Cyclin D1、p21、Bax、Bcl-2、E-cadherin和MMP-2蛋白表达水平,qRT-PCR检测lncRNA AK125512的转录水平。结果 千金子提取物可抑制MHCC97-H细胞增殖、迁移和侵袭,促进细胞凋亡,提高lncRNA AK125512的水平,降低Cyclin D1、Bcl-2和MMP-2的表达(P<0.05),促进p21、Bax和E-cadherin的表达;过表达AK125512可抑制MHCC97-H细胞增殖、迁移和侵袭,促进细胞凋亡;抑制AK125512可逆转千金子提取物对MHCC97-H细胞增殖、迁移、侵袭和凋亡的作用。结论 千金子提取物通过提高AK125512水平抑制MHCC97-H细胞的增殖、迁移和侵袭,促进细胞凋亡。千金子提取物对肝癌具有潜在治疗作用。Objective To explore the effects of Semen Euphorbiae Lathyris extract on proliferation, migration, invasion and apoptosis of hepatocellular carcinoma MHCC97-H cells and the potential mechanism.Method MHCC97-H cells were treated with 5 mg/mL, 10 mg/mL and 20 mg/mL of Semen Euphorbiae Lathyris extract respectively. The proliferation and apoptosis of MHCC97-H cells were detected by CCK-8 and flow cytometry. The migration and invasion abilities of MHCC97-H cells were determined by Transwell assay. The proteins of Cyclin D1, p21, Bax, Bcl-2, E-cadherin and MMP-2 protein levels were detected by Western blot. The transcriptional level of lncRNA AK125512 in MHCC97-H cells was measured by qRT-PCR.Result The Semen Euphorbiae Lathyris extract inhibited the proliferation, migration and invasion of MHCC97-H cells, promoted cell apoptosis, increased the level of lncRNA AK125512(P<0.05), decreased the expression of Cyclin D1, Bcl-2 and MMP-2(P<0.05), and promoted the expression of p21, Bax and E-cadherin(P<0.05). Overexpression of AK125512 inhibited the proliferation, migration and invasion of MHCC97-H cells and promoted cell apoptosis. Inhibition of AK125512 reversed the effects of Semen Euphorbiae Lathyris extract on proliferation, migration, invasion and apoptosis of MHCC97-H cells.Conclusion The Semen Euphorbiae Lathyris extract inhibits the proliferation, migration and invasion of MHCC97-H cells and promotes cell apoptosis by increasing the level of AK125512. The Semen Euphorbiae Lathyris extract has potential therapeutic effects on hepatocellular carcinoma.
