摘  要:以鼠尾藻多糖提取率为评价指标,在单因素实验的基础上,采用响应面法优化鼠尾藻多糖的超声-微波联用提取工艺,并通过测定鼠尾藻多糖对DPPH自由基、ABTS自由基的清除能力,评价其抗氧化活性.结果表明,鼠尾藻多糖的最佳提取工艺为:提取时间31min、微波功率359W、超声功率302W、液料比27∶1(mL∶g),在此条件下,鼠尾藻多糖提取率可达9.19%;鼠尾藻多糖具有一定的抗氧化活性,对ABTS自由基的清除能力(IC_(50)=0.288mg·mL^(-1))优于对DPPH自由基的(IC_(50)=0.311mg·mL^(-1)).Using the extraction rate of polysaccharides from Sargassum thunbergii as an evaluation index,we optimized the ultrasonic-microwave combined extraction process of polysaccharides from Sargassum thunbergii by response surface methodologies on the basis of single-factor experiments.Moreover,we evaluated the antioxidant activity of polysaccharides from Sargassum thunbergii by the determination of their scavenging abilities against DPPH free radicals and ABTS free radicals.The results show that the optimal extraction conditions can be determined as follows:the extraction time of 31 min,the microwave power of 359 W,the ultrasonic power of 302 W,and the liquid-solid ratio of 27∶1(mL∶g).Under above conditions,the extraction rate of polysaccharides from Sargassum thunbergii can reach 9.19%.The polysaccharides from Sargassum thunbergii has certain antioxidant activity,meanwhile,the scavenging ability of ABTS free radicals of polysaccharides from Sargassum thunbergii(IC_(50)=0.288 mg·mL^(-1))is superior to that of DPPH free radicals(IC_(50)=0.311 mg·mL^(-1)).
