摘 要:目的:探讨α-硫辛酸对帕金森病(PD)模型老年小鼠的保护作用。方法:本实验采用老年小鼠1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)-PD模型,通过长期口服给予α-硫辛酸进行干预,并通过Westernblot及免疫荧光检测不同实验组小鼠大脑黑质多巴胺神经元的损伤情况以及氧化应激途径的改变。结果:长期口服α-硫辛酸并没有显著减少正常衰老状态下的酪氨酸羟化酶神经元丢失,而在MPTP-PD模型组中长期口服α-硫辛酸能发挥抗氧化应激作用,显著保护酪氨酸羟化酶神经元免受MPTP毒物损害造成的丢失,并改善运动功能。结论:长期口服α-硫辛酸对老年PD的发生发展可能有一定的延缓、抑制作用。AIM:To explore the protective effect ofα-lipoic acid on aged mouse model of Parkinson disease(PD).METHODS:The 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)-PD model of aged mice was intervened withα-lipoic acid orally for a long time,and the damage of dopaminergic neurons in the substantia nigra and the changes of oxidative stress pathway in different experimental groups were detected by immunofluorescence technique and Western blot analysis.RESULTS:Long-term oral administration ofα-lipoic acid did not reduce the tyrosine hydroxylase neuron loss significantly in normal aging state.However,α-lipoic acid administered orally for a long time played antioxidant stress and free radical-scavenging roles in MPTP-PD model group,so as to protect against tyrosine hydroxylase neuron loss and ameliorate motor function caused by MPTP toxicity.CONCLUSION:Oral administration ofα-lipoic acid for a long time may plays a certain preventive and protective role in the occurrence and development of aged PD.
