摘 要:目的:考察奥沙利铂(oxaliplatin)联合山柰酚(kaempferol)动脉介入治疗对裸大鼠肺癌移植瘤的生长抑制和诱导凋亡作用。方法:皮下接种A549肺癌细胞,将裸大鼠分为对照组、奥沙利铂静脉注射组、奥沙利铂联合山柰酚静脉注射组,和奥沙利铂联合山柰酚动脉介入治疗组。研究考察不同给药方式对实体瘤生长、对大鼠体重的影响。利用免疫荧光染色、Hoechst染色及免疫印迹法(western blot,WB)考察瘤组织细胞凋亡水平。结果:首先相比对照组,奥沙利铂静脉注射抑制实体瘤生长,相比奥沙利铂静脉注射组,奥沙利铂静脉注射联合山柰酚对实体瘤生长没有明显抑制作用,奥沙利铂动脉介入治疗联合山柰酚显著抑制实体瘤生长。其次山柰酚能逆转奥沙利铂对大鼠的体重抑制作用。最后,静脉注射治疗下,山柰酚能促进奥沙利铂对细胞凋亡的诱导作用,而动脉介入治疗更显著地促进药物联用引起的细胞凋亡作用。结论:奥沙利铂联合山柰酚动脉介入治疗可以显著抑制裸大鼠肺癌移植瘤生长并促进细胞凋亡。Objective:To determine the antitumor effects and apoptosis induction effects of oxaliplatin combined with kaempferol via arterial chemotherapy on lung cancer cells-xenograft nude rat model.Methods:After subcutaneous inoculation of A549 cells,nude rats were divided into control group,oxaliplatin in intravenous injection group,oxaliplatin combined kaempferol in intravenous injection group and oxaliplatin combined kaempferol in arterial interventional treatment group.The effects on solid tumor growth and body weight were investigated.Immunofluorescence staining,Hoechst staining and western blot(WB)were used to investigate the apoptosis of tumor tissues.Results:Firstly,compared with control group,oxaliplatin in intravenous injection group inhibited the growth of solid tumors,which was not further inhibited by combining with kaempferol.Oxaliplatin in arterial interventional therapy combined with kaempferol significantly inhibited the growth of solid tumors.Secondly,kaempferol reversed the oxaliplatin-induced weight inhibition in rats.Finally,kaempferol could promote the induction effect of oxaliplatin on cell apoptosis,while arterial interventional therapy can significantly promote the apoptosis effect caused by drug combination.Conclusion:Oxaliplatin combined with kaempferol in arterial interventional therapy can significantly inhibit the growth and promote apoptosis of lung cancer xenografts in nude rats.
