摘  要:目的:探讨microRNA-449a(miR-449a)启动子区甲基化状态及其在子宫颈癌组织中的表达情况。方法:收集2019年01月至2021年01月我院收治的50例子宫颈癌患者的子宫颈癌组织和同期我院收治的子宫良性病变患者(50例)的宫颈组织作为对照组进行研究。甲基化特异性聚合酶链反应(methylation-specific PCR,MSP)检测两组患者宫颈组织中miR-449a启动子区甲基化状态,实时荧光定量逆转录聚合酶链反应(quantitative reverse transcription PCR,RT-qPCR)检测miR-449a在子宫颈癌患者和对照组子宫颈组织中的表达情况,同时收集两组患者临床资料进行甲基化比较。结果:子宫颈癌组织中miR-449a启动子区甲基化率为78.00%,而对照组中miR-449a启动子区甲基化率为22.00%。与对照组相比,子宫颈癌患者子宫颈癌组织中miR-449a的表达显著降低(P<0.001)。然而HPV阳性子宫颈癌患者子宫颈癌组织中miR-449a启动子区甲基化率高于HPV阴性子宫颈癌患者,且miR-449a启动子区甲基化率在FIGO I期高于FIGO II期,差异均有统计学意义。miR-449a启动子区甲基化状态与子宫颈癌患者的年龄、组织学类型、肿瘤大小、淋巴结转移和阴道入侵之间差异均无统计学意义(P>0.05)。结论:子宫颈癌中miR-449a基因的表达可能受其甲基化状态的调控,可能在子宫颈癌的发生发展中起重要作用,其甲基化状态与HPV感染和FIGO分期差异有统计学意义。Objective:To investigate the methylation status of microRNA-449a(miR-449a)promoter region and its expression in cervical cancer tissues.Methods:The cervical cancer tissues of 50 patients with cervical cancer admitted to our hospital from January 2019 to January 2021 were collected and the cervical tissues of patients with uterine benign lesions(50 cases)admitted to our hospital during the same period were collected as a control group for the study.Methylation-specific polymerase chain reaction(MSP)detected the methylation status of the miR-449a promoter region in the cervical tissues of the two groups of patients,and real-time fluorescent quantitative reverse transcription polymerase chain reaction(RT-qPCR)detected miR-449a expression in cervical cancer tissues and in the cervical tissues of the control group,and the clinical data of the two groups of patients were collected at the same time for methylation comparison.Results:The methylation rate of the miR-449a promoter region in cervical cancer tissues was 78.00%,while the methylation rate of the miR-449a promoter region in the control group was 22.00%.Compared with the control group,the expression of miR-449a in cervical cancer tissues of patients with cervical cancer was significantly reduced(P<0.001).However,the methylation rate of miR-449a promoter region in cervical cancer tissues of HPV-positive cervical cancer patients was higher than that of HPV-negative cervical cancer patients,and the methylation rate of miR-449a promoter region in FIGO I stage was higher than that in FIGO II stage,and the difference was statistically significant.There was no significant difference between the methylation status of miR-449a promoter region and the age,histological type,tumor size,lymph node metastasis and vaginal invasion of patients with cervical cancer(all P>0.05).Conclusion:The expression of miR-449a gene in cervical cancer may be regulated by its methylation status,and it may play an important role in the occurrence and development of cervical cancer,and its methy
