摘 要:目的:基于单细胞测序技术挖掘溃疡性结肠炎(UC)结肠黏膜单细胞测序数据,探讨肠上皮细胞在UC中的功能变化。方法:下载基因表达综合(GEO)数据集(GSE150115),分析该数据集中5例UC患者肠黏膜组织10 X Genomics单细胞转录组测序数据,进行细胞亚群分类、肠上皮细胞识别、细胞间差异基因筛选、基因本体(GO)功能富集和京都基因与基因组百科全书(KEGG)信号通路富集分析。结果:从10个细胞亚群中识别出cluster 5为肠上皮细胞,并筛选出细胞间差异基因。GO功能富集分析发现,在肠上皮细胞中显著差异基因包括PI3、IgG-Fc段结合蛋白(FCGBP)、紧密连接蛋白4(CLDN4)等,其差异基因主要富集于核苷酸代谢;KEGG信号通路富集分析发现,差异基因主要参与神经系统疾病。结论:肠上皮细胞的核苷酸代谢异常可能参与UC发病,同时可能增加神经系统疾病的发生风险。Objective:To explore the functional changes of intestinal epithelial cells in ulcerative colitis(UC)through dug the single cell sequencing data of UC colonic mucosa on the basis of single cell sequencing technique.Methods:The data set(GSE150115)of gene expression omnibus(GEO)was downloaded to analyze the sequencing data of 10 X Genomics single cell transcriptome of intestinal mucosa tissue of 5 UC patients in this data set.And then,the analyses were conducted for the classification of cell subset,the recognition of intestinal epithelial cell,the screening of differential genes among cells,the enrichment of gene ontology(GO)function,and the enrichment of signal paths of Kyoto encyclopedia of genes and genomes(KEGG).Results:The cluster 5 was recognized as intestinal epithelial cell from 10 cell subsets,and the differential genes among cells were screened.The analysis of GO functional enrichment indicated that the significantly differential genes included PI3,Fc fragment of IgG binding protein(FCGBP)and CLDN4 in intestinal epithelial cell.The differential genes of that mainly were enriched in nucleotide metabolism.And the analysis of KEGG signal paths enrichment indicated that the differential genes mainly were involved in the nervous system disease.Conclusion:The abnormal nucleotide metabolism of intestinal epithelial cells may be involved in the UC pathogenesis,as well as increase the risk of occurring nervous system disease.
