摘  要:目的:研究微小RNA(mi R)-451a对胰腺癌BxPc3细胞增殖、侵袭和迁移的影响。方法:体外培养BxPc3细胞,将不同浓度(50、100和200μmol/L)mi R-451a模拟物(mimic)转染至胰腺癌细胞株BxPc3中,利用荧光定量聚合酶链式反应(PCR)法检测mi R-451a的表达,MTT增殖实验检测不同浓度mi R-451a对细胞增殖的影响,细胞划痕实验检测不同浓度mi R-451a对细胞迁移能力的影响,Transwell实验检测不同浓度mi R-451a对细胞侵袭能力的影响。Western blot法检测不同浓度mi R-451a转染后BxPc3细胞p-PI3K、p-AKT、TGF-β和p-smad2/3蛋白的表达情况。结果:转染不同浓度mi R-451a mimic后,胰腺癌BxPc3细胞内mi R-451a的相对表达量明显升高,差异有统计学意义(P<0.05)。过表达mi R-451a后,BxPc3细胞的增殖能力明显减弱,细胞迁移能力明显减弱,细胞侵袭能力明显减弱,差异均有统计学意义(P<0.05)。相比于0μmol/L组,转染50、200μmol/L mi R-451a后BxPc3细胞p-PI3K、p-AKT、TGF-β和p-smad2/3蛋白表达水平明显降低(P<0.05),并且p-PI3K、p-AKT、TGF-β和p-smad2/3蛋白表达水平变化具有浓度依耐性。结论:mi R-451a能抑制胰腺癌BxPc3细胞株的增殖、迁移、侵袭能力。Objective:To study the effect of microRNA(mi R)-451a on the proliferation,invasion and migration of pancreatic cancer BxPc3 cells.Methods:BxPc3 cells were cultured in vitro,mi R-451a mimics(mimic)were transfected into pancreatic cancer cell line BxPc3 at different concentrations(50,100,and 200μmol/L).The expression of mi R-451a was detected by fluorescence quantitative polymerase chain reaction(PCR),the MTT proliferation test was used to detect the effect of different concentrations of mi R-451a on cell proliferation,and the cell scratch test was used to detect the migration ability of different concentrations of mi R-451a of cells.Transwell experiment was used to detect the influence of different concentrations of mi R-451a on the invasion ability of cells.Western blot method was used to detect the expression of p-PI3K,p-AKT,TGF-βand p-smad2/3 protein in BxPc3 cells transfected with different concentrations of mi R-451a.Results:After transfection with different concentrations of mi R-451a mimic,the relative expression of mi R-451a in pancreatic cancer BxPc3 cells was significantly increased,and the difference was statistically significant(P<0.05).After overexpression of mi R-451a,BxPc3 cell proliferation was significantly weakened,cell migration ability was significantly weakened,cell invasion ability was significantly weakened,and the differences were statistically significant(P<0.05).Compared with the 0μmol/L group,the expression levels of p-PI3K,p-AKT,TGF-βand p-smad2/3 protein in BxPc3 cells after transfection of 50 and 200μmol/L mi R-451a were significantly reduced(P<0.05),and the change levels of p-PI3K,p-AKT,TGF-βand p-smad2/3 protein are concentrationdependent.Conclusion:mi R-451a can inhibit the proliferation,migration and invasion of pancreatic cancer BxPc3 cell line.
