十一酸睾酮小鼠前列腺增生模型的建立及分析

摘  要:目的 研究以十一酸睾酮替代丙酸睾酮建立小鼠前列腺增生模型的方法,为前列腺增生动物模型的建立及药物筛选提供实验依据。方法 将50只小鼠采用随机数字表法分为正常对照组、模型一组、模型二组、模型三组和模型四组,每组10只。除正常对照组外,模型一、二、三、四组小鼠分别按不同方案肌肉注射十一酸睾酮造模,以前列腺指数及前列腺组织病理学改变作为指标,观察、比较各组造模情况,并对最佳造模方案模型小鼠给予非那雄胺片和普乐安片干预,通过模型小鼠前列腺指数及前列腺组织病理学表现的改善情况反证造模方案。结果 造模后与正常对照组比较,模型二、三组小鼠前列腺指数均明显升高,差异有统计学意义(P<0.01);模型一、二、三、四组组间比较,模型三组小鼠前列腺指数明显高于其他3组,差异均有统计学意义(P<0.05或P<0.01)。前列腺组织病理学观察结果显示,模型二、三组小鼠前列腺腺上皮细胞增生明显,呈复层或假复层,腺上皮呈乳头状,向腺腔内突出,间质血管扩张;模型一、四组小鼠前列腺腺上皮细胞多为单层,部分细胞呈立方或高柱状,少量腺上皮呈乳头状,向腺腔内突出。对最佳造模方案模型小鼠给予非那雄胺片和普乐安片干预后,小鼠前列腺指数均明显降低(P<0.05或P<0.01);前列腺组织病理学观察结果显示,前列腺腺体形态明显恢复,腺上皮内无空泡,上皮细胞胞质较为饱满,间质血管扩张有所改善。结论 推荐造模方法为:予小鼠十一酸睾酮19.2 mg/kg肌肉注射,连续2次,间隔10 d。此方法给药频次和总剂量较少,小鼠前列腺增生明显,采用临床常用药物可使增生显著改善。Objective To study the method of establishing mouse model of prostatic hyperplasia with testoste⁃rone undecanoate instead of testosterone propionate,so as to provide experimental basis for the establishment of ani⁃mal model of prostatic hyperplasia and drug screening.Methods Fifty mice were randomly divided into normal control group,model group 1,model group 2,model group 3 and model group 4,with 10 mice in each group.Except the normal control group,the mice in model group 1,2,3 and 4 were given intramuscular injection of testosterone undecanoate to establish models according to different schemes.Taking the prostate index and histopathological changes of prostate as indicators,the establishing model conditions of each group were observed and compared,and the model mice with the best modeling scheme were intervened with finasteride tablets and Pule’an Pian(普乐安片),the improvement of prostate index and histopathological manifestations of prostate in model mice disproved the modeling scheme.Results After establishing models,the prostate index of model 2 and 3 groups increased signifi⁃cantly as compared with the normal control group,and the difference was statistically significant(P<0.01).The comparison among groups 1,2,3 and 4 showed that the prostate index of mice in model 3 group was significantly higher than that of the other three groups,and the difference was statistically significant(P<0.05 or P<0.01).The histopathological observation of prostate showed that the prostatic glandular epithelial cells of mice in model 2 and model 3 proliferated significantly,presenting stratified or pseudo stratified epithelium,the glandular epithelium was papillary,protruding into the glandular cavity,and the interstitial blood vessels were dilated.Most of the prostatic glandular epithelial cells in model 1 and 4 groups were monolayer,some cells were cubic or high columnar,and a small amount of glandular epithelium was papillary and protruded into the gland cavity.After the model mice with optimal establishing mode
