缰核损毁对2型糖尿病大鼠泪膜破裂时间及泪液分泌的影响

摘　　要：目的以2型糖尿病(T2DM)大鼠为模型探讨缰核损毁对糖尿病大鼠泪膜破裂时间(BUT)及泪液分泌的影响。方法实验研究。将SD雄性大鼠随机分为对照组、T2DM、T2DM假手术组和T2DM损毁组。以高脂高糖饮食结合低剂量链脲佐菌素(STZ)诱导T2DM大鼠模型。2型糖尿病损毁组应用电损毁仪损毁双侧缰核;假手术组大鼠经历同样的手术过程,但不损毁双侧缰核。观察各组大鼠实验前后BUT、泪液分泌量和眼压的改变。结果糖尿病大鼠BUT明显缩短(P<0.001),泪液分泌量明显减少(P<0.001),眼压升高(P<0.001);缰核损毁可明显使T2DM大鼠延长BUT(P<0.001)、增加泪液分泌量(P<0.001)并降低眼压(P<0.05)。结论缰核损毁可改善T2DM大鼠干眼症状况。Objective Using rats with type 2 diabetes mellitus(T2DM)as an animal model to explore the effects of habenular nucleus(Hb)lesion on diabetic dry eye.Methods SD male rats were randomly divided into the control group,T2DM group,T2DM sham group,and T2DM lesion group.Rats with T2DM were induced on a high-fat and high-carbohydrate diet combined with low-dose streptozotocin(STZ).In the T2DM lesion group,the bilateral Hb was lesioned by an electrical current.The rats in the T2DM sham group underwent the same surgical procedure,but the Hb was not lesioned.The changes in tear film break-up time,tear secretion,and intraocular pressure were observed before and after treatments.Results The tear film break-up time of T2DM rats was significantly shortened(P<0.001),the tear secretion was significantly decreased(P<0.001),and the intraocular pressure was increased(P<0.001).The lesions of Hb could significantly prolong tear film break-up time(P<0.001),increase tear secretion(P<0.001),and decrease intraocular pressure(P<0.05)in T2DM rats.Conclusions The lesions of the Hb could improve the symptoms of dry eye in rats with T2DM.
