摘　　要：目的探讨去泛素化酶22(USP22)在食管癌组织中表达水平,分析UPS22对食管癌细胞增殖、上皮-间充质转化、侵袭和转移的影响。方法选取2021年1月到2022年1月河南省人民医院收治的67例食管癌组织和癌旁组织作为研究对象,采用组织化学分析食管癌和癌旁组织USP22表达水平。采用短发卡RNA(shRNA)技术在人食管癌细胞系SUNE-1中建立USP22敲降细胞系,并建立对照细胞系,分别命名为对照组和USP22 KD组。采用噻唑蓝(MTT)法检测两组细胞的增殖能力;采用蛋白质免疫印迹法检测两组细胞上皮-间充质转化能力;采用Transwell分析两组细胞的侵袭能力;采用异体移植瘤实验分析两组细胞的增殖和转移能力。组间比较采用t检验。结果癌旁组织中USP22表达水平(1.73±0.77)明显低于食管癌组织中USP22蛋白表达水平(2.72±0.52),差异有统计学意义(t=8.694,P<0.05)。对照组细胞48 h吸光度值(2.02±0.12)明显高于USP22 KD组细胞(1.57±0.16),差异有统计学意义(t=6.031,P<0.05)。对照组细胞在裸鼠体内生长15 d后肿瘤体积(1219.43±169.61)明显大于USP22 KD组细胞(911.71±146.17),差异有统计学意义(t=3.636,P<0.05)。USP22 KD组细胞上皮标志物E-cadherin蛋白表达水平(1.39±0.14)明显高于对照组细胞(0.52±0.14),差异有统计学意义(t=11.531,P<0.05);对照组细胞间质标志物波形蛋白(Vimentin)和Slug表达水平(1.22±0.11、1.16±0.13)明显高于USP22 KD组细胞(0.51±0.11、0.39±0.09),差异有统计学意义(t=12.010、13.171,P<0.05)。对照组细胞侵袭数量[(100.14±15.59)个]明显高于USP22 KD组细胞侵袭数量[(48.86±9.84)个],差异有统计学意义(t=7.359,P<0.05)。对照组细胞在裸鼠体内转移数量[(15.29±2.29)个]明显高于USP22 KD组细胞[(6.86±2.27)个],差异有统计学意义(t=6.921,P<0.05)。对照组细胞增强子结合蛋白4(AP4)mRNA表达水平(1.25±0.14)明显高于USP22 KD组细胞(0.40±0.13),差异有统计学意义(t=11.550,P<0.05)Objective To investigate the expression of ubiquitin specific protein 22(USP22)in esophageal carcinoma,and to analyze the effects of UPS22 on the proliferation,epithelial stromal transformation,invasion and metastasis of esophageal carcinoma cells.Methods A total of 67 cases of esophageal cancer tissues and adjacent tissues treated in our hospital from January 2021 to January 2022 were selected as the subjects.The expression of USP22 in esophageal cancer and adjacent tissues was analyzed by histochemistry.USP22 knockdown cell lines were established in human esophageal cancer cell line sune-1 by short harpin RNA(shRNA)technique,and control cell lines were established,named control group and USP22 KD group respectively.The proliferation ability of the two groups of cells was analyzed by methyl thiazolyl tetrazolium(MTT)assay.The ability of epithelial mesenchymal transformation was detected by Western blotting.The invasive ability of the two groups of cells was analyzed by Transwell.The proliferation and metastasis of the two groups of cells were analyzed by allogeneic tumor experiment.The measurement data between groups were compared by t-test.Results The expression level of USP22 in the adjacent tissues(1.73±0.77)was significantly lower than that in the esophageal cancer tissues(2.72±0.52,t=8.694,P<0.05).The 48 h light absorption value of the control group(2.02±0.12)was significantly higher than that of the USP22 KD group(1.57±0.16,t=6.031,P<0.05).The tumor volume of the control group[(1219.43±169.61)mm3]was significantly larger than that of the USP22 KD group[(911.71±146.17)mm3]after 15 days of growth in nude mice(t=3.636,P<0.05).The expression level of E-cadherin protein in USP22 KD group was significantly higher than that in control group(1.39±0.14,t=11.531,P<0.05).The expression levels of vimentin and slug in the control group(1.22±0.11,1.16±0.13)were significantly higher than those in the USP22 KD group(0.51±0.11,0.39±0.09,t=12.010,13.171,P<0.05).The number of invasive cells in the control group(10
