摘  要:目的 探讨异荭草素(Iso)对慢性不可预知应激(CUMS)抑郁模型大鼠行为学及海马神经元与炎症相关通路的作用。方法 30只雄性SD大鼠随机平均分三组(n=10):对照组(Con组,2 mg·kg^(-1)生理盐水灌胃x 6w);CUMS组:(造模后,2 mg·kg^(-1)生理盐水灌胃x 6w);治疗组(CUMS+Iso组,2 mg·kg^(-1)Iso灌胃x6w)。采用慢性不可预知应激结合孤养的方法造模,第6周及12周时进行称重。对三组大鼠进行旷场试验、糖水偏好试验、强迫游泳实验;每组大鼠中5只进行树突棘密度分析;每组大鼠中5只进行脑组织冠状切片尼氏染色;三组大鼠海马区相关炎症因子(TNF-α、IL-1β、IL-17A)进行Western blot检测。结果 与对照组相比,CUMS组大鼠体质量、旷场实验中跑动距离及站立时间、糖水偏好指数、海马CA1及CA3区尼氏染色阳性细胞数目及神经元树突棘数目均显著下降(P<0.001),CUMS组大鼠水中静止不动时间、海马中炎症因子(TNF-α、IL-1β、IL-17A)显著增加(P<0.001);而与CUMS组相比,治疗组大鼠体重、旷场实验中跑动距离及站立时间、糖水偏好指数、海马CA1及CA3区尼氏染色阳性细胞数目及神经元树突棘数目均明显增加(P<0.01),CUMS组大鼠水中静止不动时间、海马中炎症因子(TNF-α、IL-1β、IL-17A)显著减少(P <0.01)。结论 Iso可通过可通过改善CUMS大鼠海马神经元数量与结构及抑制神经炎症进而发挥其抗抑郁的作用。Objective To investigate the effects of Isoorientin(Iso) on behavioral,hippocampal neurons and inflammatory pathways in chronic unpredicted mild stress(CUMS) model rats.Methods Thirty male SD rats were randomly divided into three groups(n=10):control group(Con group,2 mg·kg^(-1)normal saline gavage x 6 w);CUMS group:(2 mg·kg^(-1) normal saline gavage x 6 w after modeling);treatment group(CUMS+Iso group,2 mg·kg^(-1)Iso gavage x 6 w).The model was made by chronic unpredictable stress combined with solitary rearing and weighed at 6 w and 12 w.Open field test,sugar water preference test and forced swimming test were carried out in three groups of rats.Density analysis of dendritic spines was performed for 5 rats in each group.Five rats in each group were treated with coronal section nissl staining.Western blot was used to detect the inflammatory factors(TNF-α,IL-1β and IL-17 A) in the hippocampus of rats in the three groups.Results Compared with the control group,body weight,running distance and standing time in open field test,sugar water preference index,the number of Nis-staining positive cells in hippocampus CA1 and CA3 and the number of dendritic spines of neurons in CUMS group were significantly decreased(P <0.001).In CUMS group,the water immovability time and inflammatory factors(TNF-α,IL-1β,IL-17 A) in hippocampus were significantly increased(P <0.001).Compared with CUMS group,body weight,running distance and standing time in open field experiment,sugar water preference index,the number of nish staining positive cells in hippocampus CA1 and CA3 and the number of dendritic spines of neurons in treatment group were significantly increased(P <0.01).In CUMS group,water immovability time and inflammatory factors(TNF-α,IL-1β,IL-17 A) in hippocampus were significantly decreased(P <0.01).Conclusion Iso plays an anti-depressive role by improving the number and structure of hippocampal neurons and inhibiting neuroinflammation in CUMS rats.
