摘  要:目的 探讨长链非编码RNA(lncRNA)HOX转录反义基因间RNA(HOTAIR)基因多态性与人乳头瘤病毒(HPV)感染女性宫颈癌易感性及临床病理参数的关系。方法 选择武汉市第一医院妇科2019年12月-2020年12月收治的HPV感染宫颈癌患者120例设为研究组,选择同期医院体检宫颈癌筛查结果HPV结果阴性女性80名为对照组,通过电子病历及问卷调查的方式收集研究对象基本资料,包括年龄、初次生育年龄、体质量指数、孕次、产次、绝经状态。采用限制性片段长度多态性聚合酶链反应检测LncRNA HOTAIR基因rs920778位点基因多态性,分析其与宫颈癌患者HPV感染的易感性及临床病理参数的关系。结果 两组研究对象分布情况在预期值与实际值之间比较差异无统计学意义,证明该研究群体具有群体代表性;研究组LncRNA HOTAIR基因rs920778位点TT基因型、T等位基因频率高于对照组,CC基因型、C等位基因低于对照组(P<0.05);Logistic回归分析结果显示携带TT基因型、T等位基因为高危型HPV感染宫颈癌发生的影响因素(P<0.05)。结论 LncRNA HOTAIR基因rs920778位点TT基因型增加本地区女性对HPV宫颈癌的易感性,但与其病情进展无关。OBJECTIVE To investigate the correlation of long non-coding RNA(lncRNA) HOX transcribed antisense intergenic RNA(HOX transcribed antisense intergenic RNA, HOTAIR) gene polymorphism, the susceptibility to cervical cancer in women with human papillomavirus(human papillomavirus, HPV) infection, and clinicopathological parameters. METHODS A total of 120 cases of cervical cancer patients with HPV infection who were admitted to the Department of Gynecology and Gynecology of Wuhan First Hospital from Dec 2019 to Dec 2020 were selected as the research group, and 80 women with negative HPV results in the physical examination of the hospital during the same period were selected as the control group. The electronic medical records and questionnaires were used to collect basic information of the research subjects, including age, age at first childbirth, body mass index, pregnancy times, parity, and menopausal status. Restriction fragment length polymorphism polymerase chain reaction was used to detect the polymorphism of LncRNA HOTAIR gene at rs920778, and analyze its relationship with the susceptibility to cervical cancer in women with HPV infection and clinicopathological parameters. RESULTS There was no significant difference between the expected value and the actual value in the distribution of the study group and the control group, indicating the representativeness of the research group. The genotype and T allele frequency of LncRNA HOTAIR gene rs920778 locus TT genotype in the research group was higher than that of the control group, and the CC genotype and C allele were lower than that of the control group. Logistic regression analysis results showed that carrying TT genotype and T allele were independent influencing factors for the occurrence of cervical cancer in women with high-risk HPV infection(P<0.05). CONCLUSION The TT genotype at rs920778 of the LncRNA HOTAIR gene significantly increased the susceptibility to cervical cancer of women with HPV infection in this region, but was not associated with the progressio
