摘  要:CCT(CO,COL和TOC1)家族基因,在植物生长发育和适应环境变化的过程中具有重要的调节作用。本研究利用生物信息学方法,对茶树基因组中鉴定到的44个CCT基因进行了基础生物信息分析,同时分析了它们在PEG诱导的干旱胁迫、盐胁迫、茉莉酸甲酯胁迫和冷处理中的转录组数据。结果表明,44个茶树CsCCT基因在茶树染色体上分布不均;根据进化关系将茶树CCT基因分为COL(Constant-like)、PRR(Pseudo-response regulator)和CMF(CCT motif family);通过基因结构与保守基序分析发现,相同亚家族的基因结构和保守结构域基本一致;顺式作用元件分析发现,激素、植物发育和胁迫响应都与顺式作用元件相关;基因表达分析显示,CsCCT基因在花和果实组织中具有较高的表达水平,且部分基因随着叶片成熟度的增加,表达水平升高;不同的CsCCT基因在PEG诱导的干旱胁迫、盐胁迫、茉莉酸甲酯胁迫和冷处理下存在差异表达,经RT-PCR分析CCT不同基因在茶树各组织表达有一定的差异和转录数据表达趋势基本一致。本研究为进一步揭示CsCCT在茶树生长发育以及抗逆响应中提供参考依据。CCT(CO,COL and TOC1)family genes have important regulatory roles in plant growth and development and adaptation to environmental changes.In this study,using bioinformatics methods,44 CCT genes identified in the tea plant genome were analyzed for basic bioinformatics,and their transcriptomes data under PEG-induced drought stress,salt stress,methyl jasmonate stress and cold treatment were also analyzed.The results showed that44 CsCCT genes were unevenly distributed on chromosomes of tea plant;tea plant CCT genes were divided into COL(Constant-like),PRR(Pseudo-response regulator)and CMF(CCT motif family)according to the evolutionary relationship.Structural and conserved motif analysis found that the gene structures and conserved domains of the same subfamily were basically the same.Cis-acting element analysis found that hormones,plant development and stress response were all related to cis-acting elements.Gene expression analysis showed that CsCCT gene expression levels were higher in flower and fruit tissues,and the expression levels of some genes increased with the increase of leaf maturity.Different CsCCT genes were differentially expressed under PEG-induced drought stress,salt stress,methyl jasmonate stress and cold treatment.RT-PCR analysis showed that the expression of different CCT genes in various tissues of tea plants had certain differences,and the expression trends of transcription data were basically the same.This study provides a reference for further revealing the role of CsCCT in tea plant growth and stress response.
