摘 要:目的:探究镁离子(Mg^(2+))对肝癌细胞的作用机制。方法:氯化镁溶液提供Mg^(2+),设置空白对照组、20 mmol/L、40 mmol/L和80 mmol/L的Mg^(2+)溶液培养基培养肝癌Huh-7细胞,检测细胞生长增殖、细胞凋亡、细胞周期,对细胞丝裂原活化蛋白激酶激酶2(MEK2)RNA产物行基因测序,Western Blot检测蛋白。结果:随着Mg^(2+)浓度升高,肿瘤细胞的生存率明显下降,细胞周期G1延长,MAP2K2 RNA上调;Western Blot检测发现在Mg^(2+)作用下Huh-7细胞的MEK2升高,与对照组比较,差异有统计学意义(P<0.05)。结论:(1)Mg^(2+)可以抑制肝癌细胞Huh-7的生长繁殖,延长肝癌细胞Huh-7的细胞周期G1;(2)MEK2有可能是Mg^(2+)抗肿瘤机制中的一个重要蛋白。Objective:To explore the mechanism of magnesium ion on hepatoma cells.Methods:magnesium chloride solution was used to provide magnesium ions,and blank control group,20 mmol/L,40 mmol/L and 80 mmol/L magnesium ion solution medium were set up to culture Huh-7 cells.The growth increment,apoptosis,cell cycle and RNA products of cells were detected by Western blot To explore its mechanism.Results:with the increase of Mg^(2+)concentration,the survival rate of tumor cells was significantly decreased,cell cycle G1 was prolonged,and MAP2K2 RNA was up-regulated;Western Blot showed that the MEK2 of Huh-7 cells increased under the treatment of Mg^(2+),and the difference was statistically significant compared with the control group(P<0.05).Conclusion:1)Mg^(2+)could inhibit the growth and reproduction of HuH-7 cells and prolong the cell cycle G1 of HuH-7 cells.2)Mitogen-activated protein kinase kinase 2 May be an important protein in the anti-tumor mechanism of Mg^(2+).
