摘  要:目的探究腺苷A1受体激动剂预处理对大鼠脑缺血再灌注损伤的保护作用,以及进一步明确其与低氧诱导因子-1α(HIF-1α)和谷胱甘肽过氧化物酶1(GPx1)之间的相关性。方法选择新乡医学院60只健康雄性SD大鼠(周龄7~9周)为研究对象,将其随机分为3组,分别为假手术组(F组,N=20只)、缺血再灌注组(IR组,N=20只)、环己基腺苷预处理组(AP组,N=20只)。其中F组、IR组方法为注入2 mL的0.9%氯化钠,AP组注入0.9%氯化钠注射液2 mL+环己基腺苷(CHA)注射液160μg/kg。各组对48 h亚组大鼠行头颅MRI检查,并对其脑组织行HE染色法。测定各组大鼠脑内不同时间点的HIF-1α、GPx1蛋白表达。结果(1)经头颅MRI检查,AP组、IR组的缺血灶体积均减小,F组未见缺血病灶。(2)HE染色,可见AP组、IR组的缺血灶处伴有脑损伤,AP组较IR组轻,而F组脑细胞正常,三组对比差异有统计学意义(P<0.05)。(3)与F组大鼠对比,其他组大鼠在6 h、24 h、48 h的HIF-1α蛋白表达水平相对较高,差异有统计学意义(P<0.05);而AP组大鼠在6 h、24 h、48 h的HIF-1α蛋白表达水平均高于IR组大鼠,差异有统计学意义(P<0.05);三组大鼠之间在2 h的HIF-1α蛋白表达水平对比差异无统计学意义(P>0.05)。(4)各时段F组大鼠的GPx1蛋白表达均高于其他两组,AP组大鼠除2 h之外,其他时段均高于IR组,差异有统计学意义(P<0.05)。结论腺苷A1受体激动剂预处理的脑保护作用较高,考虑与上调HIF-1α、GPx1蛋白的表达有关,因此有利于减轻脑缺损再灌注损伤程度,以促进预后恢复。Objective To investigate the protective effect of pretreatment with adenosine A1 receptor agonist on cerebral ischemia-reperfusion injury in rats and further clarify its relationship with hypoxia inducible factor-1α(HIF-1α)And glutathione peroxidase 1(GPx1).Methods A total of 60 healthy male SD rats(7~9 weeks old)from Xinxiang Medical College were randomly divided into 3 groups:fake operation group(F group,N=20),ischemia-reperfusion group(IR group,N=20)and cyclohexyl adenosine preconditioning group(AP group,N=20).Group F and IR were injected with 2 ml of 0.9%sodium chloride,and group AP was injected with 2 ml of 0.9%sodium chloride injection and 160 ug/kg of cyclohexyl adenosine(CHA)injection.Brain MRI was performed on 48 h subgroup rats in each group,and HE staining was performed on their brain tissues.HIF-1αand GPx1 protein expression in the brain of each group was measured at different time points.Results(1)By cranial MRI,the volume of ischemic foci in AP group and IR group decreased,and no ischemic foci were found in F group.(2)HE staining showed that the ischemic foci in AP group and IR group were accompanied with brain damage,the brain cells damaged in AP group were lighter than those in IR group,while the brain cells in F group were normal,the difference between the three groups was statistically significant(P<0.05).(3)Compared with rats in group F,the expression levels of HIF-1αprotein in other groups at 6 h,24 h and 48 h were significantly higher(P<0.05).The expression of HIF-1αprotein in AP group was significantly higher than that in IR group at 6 h,24 h and 48 h(P<0.05);There was no significant difference in HIF-1αprotein expression between the three groups at 2 hours(P>0.05).(4)The GPx1 protein expression of rats in group F was higher than that of the other two groups at each time.Except for 2 h,the GPx1 protein expression of rats in group AP was higher than that in group IR,with a significant difference(P<0.05).Conclusion Pretreatment with adenosine A1 receptor agonist has a higher protective e
