摘 要:目的:研究嗅三针对蛛网膜下腔出血并发认知障碍模型大鼠的干预作用及对海马神经元Caspase-3、PI3K、AKT mRNA和蛋白表达的影响。方法:取清洁级雄性SD大鼠60只,随机分为空白组、模型组、嗅三针组、尼莫地平组以及假针刺组,每组12只。各组大鼠干预后,采用荧光定量PCR检测大鼠海马区Caspase-3、PI3K、AKT mRNA表达,Western blot技术检测Caspase-3、PI3K、AKT蛋白表达量。结果:与模型组比较,嗅三针组和尼莫地平组海马中PI3K、AKT mRNA表达量均有不同程度升高,且嗅三针组高于尼莫地平组,差异有统计学意义(均P<0.05),嗅三针组和尼莫地平组Caspase-3 mRNA表达均低,差异有统计学意义(均P<0.05)。与模型组比较,尼莫地平组及嗅三针组PI3K、AKT蛋白的表达均升高,Caspase-3蛋白表达降低,差异有统计学意义(均P<0.05)。结论:嗅三针对蛛网膜下腔出血并发认知障碍大鼠的神经保护作用机制可能是通过激活大鼠脑组织的PI3K/AKT信号通路,抑制Caspase-3蛋白表达及细胞凋亡,减轻神经组织炎性损伤。Objective:To study the intervention effect of Xiusan Zhen on subarachnoid hemorrhage concurrent cognitive impairment model rats and the effects on mRNA and protein expressions of Caspase-3,PI3K and AKT in hippocampal neurons.Methods:60 clean grade male SD rats were randomly divided into blank group,model group,sniffing group,nimodipine group and Xiusan Zhen group,12 rats in each group.Each group after corresponding intervention,the mRNA expression levels of Caspase-3,PI3K and AKT in the hippocampus of rats were detected by fluorescence quantitative PCR,protein expression levels of Caspase-3,PI3K and AKT were detected by Western blot.Results:Compared with the model group,the mRNA expression of PI3K and AKT in the hippocampus of the Xiusan Zhen group and the nimodipine group were both increased to different degrees,and the Xiusan Zhen group was higher than the nimodipine group,difference statistically significant(all P<0.05).The mRNA expression of Caspase-3 in Xiusan Zhen group and the nimodipine group was significantly lower,and difference statistically significant(all P<0.05).Compared with the model group,the protein expression of PI3K and AKT were significantly higher in the nimodipine group and the Caspase-3 protein expression was significantly lower than those in the nimodipine group,difference statistically significant(all P<0.05).Conclusion:The main mechanism of neuroprotective effect of Xiusan Zhen on cognitive impairment in rats with subarachnoid hemorrhage may be through activation of PI3K/AKT signaling pathway and inhibition of Caspase-3 protein expression in rat brain tissue and apoptosis,reducing inflammatory damage in neural tissue.
