摘  要:目的 探究辣木叶通过抗氧化作用改善血管性痴呆(VD)小鼠学习记忆的机制。方法 通过Himori法制备VD小鼠模型,90只VD模型小鼠随机分为VD组、辣木叶提取液组和辣木叶发酵物组(n=30)。30只健康小鼠作为对照组。辣木叶提取液和发酵物灌胃干预(400 mg·kg^(-1))。28 d后进行水迷宫实验分析学习记忆功能。比较各组海马组织损伤、海马神经元凋亡、氧化应激指标、PI3K/AKT信号通路的水平。结果 四组小鼠上述各指标比较差异显著(P<0.05)。VD组的目标象限停留时间、穿越平台次数、超氧化物歧化酶(SOD)、磷脂酰肌醇3激酶(PI3K)、蛋白激酶B (AKT) mRNA和蛋白水平显著低于对照组,而海马神经元凋亡、脂质过氧化产物丙二醛(MDA)水平显著高于对照组(P<0.05)。辣木叶提取液组的目标象限停留时间、穿越平台次数、SOD、PI3K、AKT mRNA和蛋白水平显著高于VD组,而海马神经元凋亡、MDA水平显著低于VD组(P<0.05)。辣木叶发酵物组的目标象限停留时间、穿越平台次数、SOD、PI3K、AKT mRNA和蛋白水平显著高于辣木叶提取液组,而海马神经元凋亡、MDA水平显著低于辣木叶提取液组(P<0.05)。结论 辣木叶提取液和发酵物能够保护VD小鼠学习记忆能力,促进PI3K/AKT通路并缓解氧化应激反应和海马神经元凋亡,辣木叶发酵物缓解VD的作用优于辣木叶提取液。Objective To explore the mechanism of Moringa oleifera leaf s improving learning and memory in vascular dementia(VD) mice through antioxidant effects.Methods VD mouse model was prepared by Himori method.Ninety VD model mice were randomly divided into VD group,Moringa oleifera leaf extract group and Moringa oleifera leaf fermentation group(n=30).Thirty healthy mice were used as the control group.Moringa oleifera leaf extract and fermentation were intervened by gavage(400 mg·kg^(-1)).After 28 d,the learning and memory function was analyzed through the water maze experiment.The levels of hippocampal tissue damage,hippocampal neuronal apoptosis,oxidative stress indicators,and PI3 K/AKT signaling pathways in each group were compared.Results The above indicators of the four groups of mice were significantly different(P<0.05).The target quadrant stay time,the number of crossing platforms,superoxide dismutase(SOD),phosphatidylinositol 3 kinase(PI3 K),protein kinase B(AKT)mRNA and protein levels in the VD group were significantly lower than those in the control group,while the hippocampal neuronal apoptosis and lipid peroxidation product malondialdehyde(MDA) levels were significantly higher than those in the control group(P<0.05).Moringa oleifera leaf’s target quadrant residence time,number of crossing platforms,SOD,PI3 K,AKT mRNA and protein levels were significantly higher than those of the VD group,while hippocampal neuronal apoptosis and MDA levels were significantly lower than those of the VD group(P<0.05).The target quadrant residence time,number of crossing platforms,SOD,PI3 K,and AKT mRNA and protein levels of the Moringa oleifera leaf fermentation group were significantly higher than those of the Moringa oleifera leaf extract group,while hippocampal neuronal apoptosis and MDA levels were significantly lower than those of Moringa oleifera leaf extract group(P<0.05).Conclusion Moringa oleifera leaf extract and fermentation can protect the learning and memory ability of VD mice,promote the PI3 K/AKT pathway and r
