摘 要:本研究以低温锡箔烘焙裸仁南瓜籽为对象,采用超声辅助溶剂法提取南瓜籽油,运用Box-Behnken-Matlab分析法对南瓜籽油进行基于亚油酸(LA)相对含量(Y_(1))和粗油得率(Y_(2))的提取工艺优化。结果表明:采用Box-Behnken-Matlab分析法得到南瓜籽粗油最优提取工艺参数为:超声时间15 min,料液比1∶38.65,提取温度25.75℃,提取时间2.7 h,此时,Y_(1)可达(48.23±0.11)%,Y_(2)为(24.83±0.12)%;当超声时间15 min,提取时间的范围为2.5~3 h,料液比1∶34~1∶39,提取温度22~27℃时,在此工艺参数区间Y_(1)和Y_(2)可同时取得较大理论值,此时,粗油中总酚含量为(968.71±13.96)mg GAE/kg。研究表明,Box-Behnken-Matlab分析法不仅可以得到高LA相对含量的南瓜籽油最优工艺参数,也可以在兼顾LA相对含量和粗油得率的情况得到较优工艺参数范围,为高活性南瓜籽油工业化提取提供了参考。In the present study,the pumpkin seed oil was extracted by ultrasonic-assisted solvent method,and the extraction process was optimized based on the relative content of linoleic acid(LA)(Y_(1))and crude oil yield(Y_(2))by Box-Behnken-Matlab analysis.The results indicated that the ultrasonic time had no significant effect on the Y_(1) of pumpkin seed oil(P>0.05),while the solid-liquid ratio with petroleum ether as the solvent,extraction temperature and extraction time had significant effect on the Y_(1) of pumpkin seed oil(P<0.05).The optimal extraction parameters of pumpkin seed were obtained by Box-Behnken-Matlab analysis:the ultrasonic time was 15 min,the solid-liquid ratio was 1∶38.65,the extraction temperature was 25.75℃,and the extraction time was 2.7 h.Under the optimal conditions,Y_(1) and Y_(2) could reach(48.23±0.11)%and(24.83±0.12)%,respectively.When the extraction time was 2.5~3 h,the solid-liquid ratio was 1∶(34~39),and the extraction temperature was 22~27℃,larger theoretical values of Y_(1) and Y_(2) could be obtained at the same time in this process parameter range.Under these conditions,the total phenolic content in crude oil was(968.71±13.96)mg GAE/kg.The results indicated that Box-Behnken-Matlab analysis method could not only obtain the optimal process parameters of pumpkin seed oil with high relative LA content,but also obtain the optimal process parameters range when considering the relative LA content and crude oil yield,which provided a reference for the industrial extraction of high-activity pumpkin seed oil.
