摘 要:目的探讨蛇葡萄素(AMP)对人膀胱癌J82细胞增殖和凋亡的影响及相关作用机制。方法配置不同浓度(40.00、44.74、50.04、55.96、62.59、70.00μg/ml)的AMP作用于人膀胱癌J82细胞,分别培养48、72 h,使用噻唑蓝检测不同浓度的AMP对人膀胱癌J82细胞增殖能力的影响,流式细胞术检测不同浓度(40.00、50.04、62.59μg/ml)的AMP对人膀胱癌J82细胞凋亡能力的影响,透射电镜观察细胞形态,蛋白质印迹法(Western blot)检测磷酸化的B细胞淋巴瘤/白血病-2(p-Bcl-2)、p-Bad的相对表达量。结果不同浓度AMP处理人膀胱癌J82细胞48、72 h,抑制率(IR)随AMP浓度升高、作用时间的延长逐渐升高,具有明显的浓度依赖性和时间依赖性。AMP处理48 h的半数抑制浓度(IC50)明显高于AMP处理72 h细胞,差异有统计学意义(P﹤0.01)。40.00、50.04、62.59μg/ml AMP组和阳性对照组人膀胱癌J82细胞的总凋亡率均高于阴性对照组,40.00、50.04、62.59μg/ml AMP组人膀胱癌J82细胞的总凋亡率均高于阳性对照组,差异均有统计学意义(P﹤0.05)。随着AMP浓度的增加,J82细胞存活细胞率逐渐降低,早期凋亡率先上升后下降,晚期凋亡率和总凋亡率均逐渐升高。40μg/ml的AMP作用于人膀胱癌J82细胞48 h后,透射电镜下可见凋亡细胞体积变小,细胞质浓缩,出现凋亡小体。作用24、48 h,70μg/ml的AMP处理的人膀胱癌J82细胞凋亡抑制蛋白p-Bad、p-Bcl-2相对表达量均低于处理12 h时(P﹤0.05)。结论AMP可有效抑制人膀胱癌J82细胞的增殖,促进其凋亡,诱导机制可能与其能够下调抗细胞凋亡蛋白p-Bad、p-Bcl-2的表达有关。Objective To investigate the effect and related mechanism of ampelopsin(AMP) on the proliferation and apoptosis of human bladder cancer J82 cells.Method Different concentrations(40.00,44.74,50.04,55.96,62.59,70.00 μg/ml) of AMP were prepared to treat with human bladder cancer J82 cells.They were cultured for 48 and 72 h,respectively.The effect of different concentrations of AMP on the proliferation of human bladder cancer J82 cells was detected by methyl thiazolyl terazolium,and the effect of different concentrations(40.00,50.04,62.59 μg/ml) of AMP on the apoptosis of human bladder cancer J82 cells were detected by flow cytometry.In addition,cell morphology was observed by transmission electron microscopy.The relative expression levels of phosphorylated B cell lymphoma/leukemia-2(p-Bcl-2) and p-Bad were detected by Western blot.Result Human bladder cancer J82 cells were treated with different concentrations of AMP for 48 and 72 h,inhibition rate(IR) increased with the increase of AMP concentration and the prolongation of action time,and the index had significant concentration and time dependence.The half inhibitory concentration(IC50) of cells treated with AMP for 48 h was significantly higher than that of cells treated with AMP for 72 h,and the difference was statistically significant(P<0.01).The total apoptosis rate of human bladder cancer J82 cells in the40.00,50.04,62.59 μg/ml AMP groups and the positive control group were higher than those in the negative control group,and the rate of human bladder cancer J82 cells in the 40.00,50.04,and 62.59 μg/ml AMP groups were higher than those in the positive control group,and the differences were statistically significant(P<0.05).The survival rate of J82cells gradually decreased,while the early apoptosis rate first increased and then decreased,and the late apoptosis rate and total apoptosis rate gradually increased with the increase of AMP concentration.After 40 μg/ml of AMP was applied to human bladder cancer J82 cells for 48 hours,the volume of apoptotic cells
