摘  要:目的:观察5首活血化瘀方对血瘀证新西兰兔血脂代谢、肝脏组织及三磷酸腺苷结合盒转运体A1(ABCA1)、过氧化物酶体增殖物激活型受体γ(PPARγ)表达的影响,对比其作用差异,为临床选方用药提供实验室依据。方法:70只新西兰兔随机分为正常组(10只)和造模组(60只),造模组采用“饥饿+高脂饲料+肾上腺素”方法建立血瘀证模型,造模成功后随机分为模型组、血府组(血府逐瘀汤3.55 g·^(-1)·d^(-1))、丹参组(丹参饮1.962 g·^(-1)·d^(-1))、失笑组(失笑散0.56 g·kg^(-1)·d^(-1))、活络组(活络效灵汤2.80 g·^(-1)·d^(-1))、桃红组(桃红四物汤2.66 g·^(-1)·d^(-1)),各复方组灌胃相应复方,正常组和模型组灌胃同等剂量蒸馏水,连续灌胃30 d。治疗结束后腹主动脉取血,检测血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、载脂蛋白A1(ApoA1)含量;苏木素-伊红(HE)染色观察肝脏组织病理形态学变化,实时荧光定量聚合酶链式反应(Real-time PCR)检测肝脏组织ABCA1、PPARγmRNA表达,蛋白免疫印迹法(Western blot)检测ABCA1、PPARγ蛋白质表达。结果:与正常组比较,模型组HDL-C、LDL-C、TG、TC水平、PPARγmRNA及蛋白质水平升高(P<0.01),ApoA1水平降低(P<0.05),ABCA1 mRNA及蛋白质水平降低(P<0.01);与模型组比较,5首活血化瘀方HDL-C水平均降低(P<0.05,P<0.01),血府组与失笑组TG水平降低(P<0.05),失笑组LDL-C、TC水平降低(P<0.05),活络组、桃红组ApoA1水平升高(P<0.05,P<0.01);血府组、失笑组、活络组、桃红组ABCA1 mRNA水平均升高(P<0.01),5首活血化瘀方均可升高ABCA1蛋白质水平(P<0.05,P<0.01);5首活血化瘀方PPARγmRNA水平均降低(P<0.01),血府组、失笑组、活络组、桃红组PPARγ蛋白质水平均降低(P<0.01)。结论:5首活血化瘀方对血脂异常有一定的调治作用,其可能是通过增加ApoA1、ABCA1的表达促进HDL-C的生成及加强功能异常HObjective:To observe the effects of five Huoxue Huayu prescriptions on blood lipid metabolism,liver tissue and adenosine triphosphate binding cassette transporter A1(ABCA1)and peroxisome proliferator-activated receptor γ(PPARγ)expression in New Zealand rabbits with blood stasis syndrome,and to compare their differences in order to provide laboratory evidence for clinical selection of prescriptions and drugs.Method:Seventy New Zealand rabbits were randomly divided into normal group(n=10)and model group(n=60).The blood stasis syndrome was modeled by the method of starvation+high-fat feed+adrenaline.After the models were successfully established,they were randomly divided into Xuefu Zhuyutang(3.55 g·^(-1)·d^(-1))group,Danshenyin(1.962 g·^(-1)·d^(-1)) group,Shixiaosan(0.56 g·^(-1)·d^(-1)) group,Huoluo Xiaolingdan(2.80 g·^(-1)·d^(-1)) group,and Taohong Siwutang(2.66 g·^(-1)·d^(-1)) group,and were given corresponding compound prescriptions by gavage.The normal group and model group were given the same dose of distilled water.After the treatment of 30 consecutive days,blood was taken from the abdominal aorta to detect the content of total cholesterol(TC),triglycerides(TG),high-density lipoprotein cholesterol(HDL-C),low-density lipoprotein cholesterol(LDL-C)and apolipoprotein A1(ApoA1).Hematoxylin-eosin(HE)staining was used to observe the changes in liver tissue.Real-time polymerase chain reaction(Real-time PCR)and Western blot were used to detect the mRNA and protein expression of ABCA1 and PPARγ in liver tissue,respectively.Result:Compared with the conditions in the normal group,increased mRNA and protein levels of HDL-C,LDL-C,TG,TC,and PPARγ(P<0.01),decreased ApoA1 level(P<0.05)and decreased mRNA and protein levels of ABCA1(P<0.01)were found in the model group.Compared with the conditions in the model group,the HDL-C level in the five Huoxue Huayu prescriptions was lowered(P<0.05),and lowered TG level in Xuefu Zhuyutang group and Shixiaosan group(P<0.05),decreased LDL-C and TC levels in Shixiaosan group(
