摘 要:目的探讨华中地区成年汉族人群肥胖与PYY受体基因rs1047214位点多态性的关联性。方法选择华中地区成年汉族肥胖病例400名,同时选取该地区年龄±3岁、性别相同的非肥胖人群作为对照,并提取研究对象的10ml静脉血用于血液生化指标检测、提取DNA和rs1047214位点多态性检测。结果检出PYY受体基因rs1047214位点基因型CC在肥胖组中的分布频率明显高于对照组,差异具有统计学意义(P<0.05),调整体力活动、TG等混杂因素候,基因型CC的OR(95%CI)为2.73(1.13-6.61)。对不同基因型生化指标分析,TC+CC、TC、TT、TC+TT基因型的TG和HDL-C指标均值分布差异具有统计学意义(P均<0.05),TC+TT基因型的血糖和胆固醇(TC)指标均值分布差异具有统计学意义(P均<0.05)。结论PYY受体基因rs1047214位点基因型CC可能与汉族人群肥胖的发生存在关联,且基因型TC+TT可能与血糖和胆固醇(TC)的升高可能存在关联,基因型TC+CC、TC、TT、TC+TT可能与TG升高和HDL-C降低存在关联。Objective To investigate the association between obesity and rs1047214polymorphism of PYY receptor gene in han adults in central China.Methods A total of 400obese han adults in central China were selected,and nonobese people of the same sex and±3years old were selected as the control group.10ml venous blood of the subjects was extracted for blood biochemical index detection,DNA extraction and rs1047214polymorphism detection.Results The distribution frequency of genotype CC at rs1047214of PYY receptor gene was significantly higher in the obesity group than in the control group,with statistically significant difference(P<0.05).After adjusting for confounding factors such as physical activity and TG,the OR(95%CI)of genotype CC was 2.73(1.13-6.61).The analysis results of biochemical indexes of different genotypes showed that the mean distribution differences of TG and hdl-c indexes of TC+CC,TC,TT and TC+TT genotypes were statistically significant(all P<0.05),while the mean distribution differences of blood glucose and cholesterol(TC)indexes of TC+TT genotypes were statistically significant(all P<0.05).Conclusion Genotype CC at rs1047214of PYY receptor gene may be associated with obesity in han population,and genotype TC+TT may be associated with elevated blood glucose and TC,while genotype TC+CC,TC,TT and TC+TT may be associated with elevated TG and decreased HDL-C.
